Influence of Stem Cell Factor Combined with Interleukin-3 on Expansion in Vitro of Murine Bone Marrow Cells and its Hematopoietic Reconstitution Capacity

Abstract: Investigation was carried out on the effects of stem cell factor (SCF) in combination with interleukin-3(IL-3) on expansion of mice 5FU treated 3 days′ bone marrow cells(d3-5FU-BMC )in vitro using the method of colony culture and hematopoietic recovery in lethally irradiated mice transplanted with the expanded cells .The result demonstrated that the number of granulocyte-macrophage colony forming units(GM-CFU) and high proliferative potential colony forming units(HHP-CFU) increased (24.2±6.1) fold and (1.5±0.3) fold(P<0.05) respectively compared with the control group when SCF was used alone.When SCF combined with IL-3,the number of GM-CFU and HHP-CFU was (30.5±9.5) (P>0.05 compared with SCF alone)and (13.6±4.5) times of that of the control group(P<0.05 compared with SCF alone). The result of mice bone marrow cells transplantation experiment demonstrated that the number of recipients’ peripheral blood cells which received lethally irradiation transplanted with expanded d3-5FU-BMC with SCF,SCF+IL-3 recovered earlier by 1-5 days than the control group.Not only the number of the peripheral blood cells of SCF+IL-3 group recovered the earliest,but also the number of transplanted cells were reduced more than 10 folds. The result suggested that SCF had an effect on expansion of mice bone marrow cells in vitro.There was synergistic effect between SCF and IL-3.The expanded bone marrow cells could accelerate early hematopoietic reconstitution and reduce the number of transplanted cells.
