Studies of the Effect of α,α'-dithiobis [β-2-furan)acrylic acid] on Binding of Zinc to HSA by Gel Chromatography

Abstract:

In this paper,the effect of α,α′-dithiobis[β-(2-furan)acrylicacid](DTFA)on binding Zn to HSA by gel chromatography wasstudied.The amounts of HSA bound Zn,the molar ratio of Zn to DTFAand fluorescence spectrum of HSA were determind.It was found that:1.The DTFA could markedly increase the amount of HSA boundZn.In systems of HSA,Zn and DTFA the amount of Zn-HSA was 66.40%and the amount of Zn-HSA only 38.28% in the systems of Zn and HSA.The results obtained in the present investigation were consistent withthe animal experiment.2.The principal peak in the fluorescence spectrum of HSA was 345nm on excitation wave length 295 nm.Asimilarity in wave length of thefluorescence peak of HSA-Zn-DTFA systems was observed.Theseobservations suggestsd that mechanism of DTFA induced increase inserum zinc concentration involved the formation of a ternary complex ofAlbumin,Zinc and DTFA.
