A Study of ENA Antibodies with Counterimmuno-electrophoresis(CIE),One and Two Dimensional Electrophoresis and Immunoblotting(1D-IB,2D-IB)

Abstract: Twenty SLE sera were examined for ENA autoantibodies with calf thymus and human spleen extracts,and HEP-2 cell lysate as antigens in CIE,and HEP-2 lysate as antigen in 1D-IB and 2D-IB.1D-IB was more sensitive than CIE for the detection of Sm antibody only. 2D-IB could effectively detect SS-A and SS-B antibodies and analyze their fine specificities; it could demonstrate RNP antibodies reacting with 68 kD and 20 kD polypeptides,but had a low detection rate for Sm antibody(3 positives out of 20 sera).The use of HEP-2 cell lysate as antigen in CIE yielded a low detection rate for SS-B antibody(2/20).
