The Establishment of Axenic Culture of a Fujian Strain of Giardia Lamblia

Abstract:

Cysts of G. lamblia obtained and purified form a 6-year-old boy living in Fujian province south-easthern China,were inoculated orally into the suckling gerbils(Meriones unguiculatus).Trophozoites of this organism isolated aseptically from the upper parts of the intestines of the infected animals were seeded into modified TYI-S-33 medium and incubated at 37 ℃.72 h after the cultivation,the organisms grew luxuriantly and formed a monolayer on the inner surface of the culture tube.During the period of the subcultures,the cells still grew well.No baterial(s)and(or)other micrcoganism(s)were found by inoculating aliquots of the culture into beef broth for cultivation at 37℃ for 5 days.After the organisms were cryopreserved in liguid nitrogen for 5months,the viability of the thrawed organisms ranged from 70%~75%.The present strain of the axenic culture of G. lamblia was designated as FUJI/92/DPCIM/1.
