An Approach to the Pathologic Diagnosis of Early Myocardial Ischemia ──immuno-histochemical staining method for creatine kinase

Abstract:

Pathologic diagnosis of early myocardial ischemia is a very important problem. We tried to research the specificity and sensitivity of creatine kinase immunohistochemical stain for the diagnosis of early myocardial ischemia. At first,the pure antigen was extracted from the skeletal muscles of SD rats. The rabbit was immunized and anti-rat antibody was obtained.The left coronary artery of female SDrat was ligated in order to make the myocardium ischemic 20, 40, 60, 120 and 140 min after ligation, the heart was taken out, the myocasdial specimens were embeded in paraffin and their sections were stained according to PAP method. The results showed that discolouration of the ischemic myocardium accurred if ischemic time was more than 40 min. So,we conclude that the creatine kinase PAPstaining method is an effective diagnostic method and has higher specificity and sensitivity for early myocardial ischemia.
