Restriction Fragment Length Differences (RFLDs) Analysis of DNA Isolates of Trichinella spiralis from China

Abstract:

The restriction fragment length differences (RFLDs) of 4 domestic isolates of Trichinella spiralis from Yunnan, Henan, Shenyang, Changchun of China, were analysed. Trichinella spiralis of Yunnan, Henan, Shenyang were isolated from pig, while Trichinella spiralis of Changchun was isolated from dog. Total genomic DNA of muscle larve of each isolates was extracted with phonol/ethanol methods and digested separately with 10 restriction enzymes (BamHI,Dral,EcoRI,Hae Ⅲ, Hind Ⅲ, Hpa Ⅱ, Pst I, Pvu Ⅱ, XbaⅠ, XhoⅠ). All the DNA digested with enzymes were then carried on agarose gel electrophoresis for RFLDs analysis.The results shown that pattern differences were observed between isolates from pig and dog,and differences of host was useful more than differences of geography and climate in the taxonomy of Trichinella spiralis.
