Abstract: The effect of recombinant human tumor necrosis factor-a(rhTNFα)on leukemic cells was investigeted. The suppressive effect of rhTNFon HL60 and K562 leukemic cells was time and dose dependent. After incubation for 24 hours, the suppressive effect of rhTNFαon HL60 and K562 cells should be above 1μg at 2×105 leukemic cells. Our work indicated that rhTNFαwhen used conjointly with Vp16 exerted a synergetic action on inhibiting the growthof leukemic cells and that these results of two methods were generally the same. The suppressive effect of rhTNFαon colony forming unit-leukemia(CFU-L)in two patiensts with acute myelogenous leukemia was shown. These observations suggested that the rhTNFαcould be used to treat the patient with acute myelogenous leukemia.

Key words: rhTNFα(recobinant human tumor necrosisfactor-a), leukemia, MTT(tetrazolium-based semiautomated colorimtric assay)
