Separation and Determination of Eicosapentaenoic Acid and Docosahexaenoic Acid in Fish Oil by High Performance Liquid Chromatography

Abstract: The method of high performance liquid chromatography analysis for eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in the anchovy oil was reported.The sample was determined with HPLC system of model Waters and μ-Bondpark C18 colume (30cm×4 mm i.d).The EPAand DHA in fish oil was saponified with 0.5 mol/L KOH-C2H5OH,esterified with triethylamine-ω-Bromoacetopnenone, and inject directly into the system of HPLC.UVdetection was performed at 254 nm,with the mobile phase of Methanol-water(88:12).The contents of the EPA and DHA in the sample were determined by standard curve.The recovery rates of EPA and DHA were 93.0%~98.0% and 93.0%~100.0% respectively.The relative standard deviations of EPA and DHA were 0.41%~0.86% and 0.67%~1.20% (n=7).This method is simpie,rapid and with good repeatability.
