Abstract:

For efficent expression of the tyrosine hydroxylase (TH) gene, the molecular cloning was used to construct a vector containing the TH gene and a promoter, the herpes simplex virus type Ⅰ-thymidine kinase (HSV1-TK).The promoter was from the plasmid,pHSV-106 and the TH gene from the plasmid PKSTH.The newly reconstructed plasmid PHSV1TH was identified by analysis of restriction endonuclease and \.The results indicated that the HSV1-TH plasmid would be a useful tool for gene therapy of Parkinson' s disease.

Key words: tyrosine hydroxylase gene, herpes simplex virus type Ⅰ-thymidine kinase gene, DNA plasmid reconstruction, PHSV1 TH plasmid
