Role of Endothelin in Rat Abdominal Aortic Thrombolysis

Abstract: Using a rat abdominal aortic thrombosis model induced by directcurrent-ferric trichloride method, we found that plasma ET level was 3.7 times as baseline after lysing the thrombus with urokinase 250000 units per kilogram, while 5 microgram per kilogram of exogenous ET prolonged the time of thrombolysis,which was 115% of baseline,meanwhile plasma ET was 6 times as baseline.Incubation of rat aortic strip in vitro showed that 20 mmol/L of taurine had no effect on base ET release,but it inhibited the stimulation of thrombin and A23187 to ET in a dose dependent way.Plasma ET had 17% and 40% of decrease in vivo after injection of taurine,and it required a 45% shorter time to lyse the thrombus.We conclude that ET inhibits thrombolysis,its abnormally increasing can be weakened by taurine which is benificial to thrombolysis.
