Abstract:
epatocyte transplantation had been used for hepatic support and gene therapy. Except for in spleen and portal vein,hepatocytes are difficult to live and maintain hepatic tissue speciality.This study dealed with attaching hepatocytes to “polyglycolic acid-Collagen carrier" in cell culture and then implanting this polymer cell scaffold into animal's omentum: multiple fibers of polymer were fashioned from absorbable polyglycolic acid suture and then coated with type Ⅰcollagen which was prepared from rat tail tendons.Co-cultured hepatocytes were isolated from SDrats with this polymer scaffold for 96 hours and then implanted into omentum of Wistar rats which undertook partial hepatectomy. Successful engraftment occurred in the omentum were observed 14,28,60 days after transplantation,which showed viability of hepatocytes with mitotic figures.PASstain showed rich glycogen within the cytoplasm of the implanted hepatocytes.This method of hepatocyte transplantation is an alterative to current way.
