Establishment of Rat Model of Vascular Depression

Abstract: Objective To explore the establishment of animal model of arteriosclerosis combined with depression.Methods Arteriosclerosis combined with depression model was established by injecting intramuscularly with vitamin D3 300 000U/kg for 3 days,feeding with high lipid feed for 9 weeks and followed by chronic unpredictable mild stress(CUMS) for 3 weeks.The model was evaluated with weight,blood fat,activity and interest(Open-field test) and hedonia(test of 1% sucrose solution and pure water intake).Results Comparing with the control group,the model group has four results.1.the weight decreased markedly(t=-5.591,P<0.001).2.CHO and LDL increased markedly(t=5.31,P<0.001;t=5.943,P<0.001),the HDL decreased markedly(t=-2.118,P<0.05).3.the vertical scores,horizontal scores and total scores in Open-field test decreased markedly(t=-3.989,P<0.01;t=-6.368,P<0.001;t=-6.254,P<0.001);4.Intake of 1% sucrose solution percentage decreased markedly(t=-2.296,P<0.05) and the pure water intake increased markedly(t=2.397,P<0.05).Conclusion This model was characterized with weight,HDL of blood,activity,interest and hedonia decreased obviously and CHO,LDL of blood and pure water intake increased markedly,which has the symptoms of both hyperlipidemia and depression.So,the model,which established by injecting intramuscularly with vitamin,feeding with high lipid feed and followed by chronic unpredictable mild stress(CUMS),is similar to clinical manifestation of vascular depression in human.It is an appropriate model of vascular depression.
