Effect of ATP on Human Nasal Ciliary Beat Frequency

Abstract: Objective To investigate the effects of ATP on human nasal cilia movement. Methods Ciliary beat frequency(CBF) of cultured human nasal epithelial cells was measured by high-speed digital microscopy in ATP solution of different concentrations. Results CBF of cultured nasal epithelial cells exposed to HBSS showed no significant changes in 3 minutes(P>0.05); However in 1, 10 and 100 μmol/L ATP solution, CBF increased significantly in 10 seconds and reached the apex, then it showed a sustained elevation in 3 minutes. In 1 μmol/L ATP solution, CBF increased significantly from(9.2±1.8)Hz to(10.3±2.0)Hz(n=10)(P<0.05). Similarly in 10 μmol/L ATP and 100 μmol/L ATP solution, CBF increased significantly when compared with samples in HBSS. Conclusion When 1, 10 and 100 μmol/L ATP were added to the chamber, there were rapid increase in ciliary activity. In addition, there is concentration-dependent relation between CBF and ATP.
