Electron Microscopic Observations of the Endoplasmic Reticulum in Whole-Mount Cultured Cells

Abstract:

We have employed the technique of whole-mount cultured cell fixed with potassium permanganate to examine the ultrastructure of the endoplasmic reticulum(ER)in cells of different cell lines(Hela,LA795and 2BS).and revealed a continuous ER network consisting of interconnecting tubules extending throughout the cytoplasm.We have also examined the ultrastructure and threedimensional relationships of mitochondria.The cristae of mitochondria is clearly seen in Hela cells fixed with potassium permanganate for 7 min at room temperatrue. The results suggest that this method is very useful for direct examination of the ultrastructure and distribution of membrnae organelles.
