Inhibitory Effect of Tenuigenin on Beta Amyloid Protein Production of SH-SY5Y Cells Transfected with APP695 cDNA

Abstract:

Objective To investigate the inhibitory effect of tenuigenin on beta amyloid protein(Aβ) which caused Alzheimer's Disease(AD).Methods Western blotting was used to test the Aβ and C99 levels of cells; MTT and LDH was used to test the neurotoxicity of tenuigenin to the cells.Results Tenuigenin can inhibit the production of beta amyloid protein of SH-SY5Y APP695 cells, didn't have neurotoxicity to cells; it can decrease Aβ and C99 levels of SH-SY5Y APP695 cells, but it didn't decrease Aβ and C99 levels of SH-SY5Y SPA4CT cells.Conclusion Tenuigenin can inhibit the production of beta amyloid protein of SH-SY5Y APP695 cells, it's mechanism may come through affecting the β-secretase proteolytic activities.
