The Changes of Toll-like Receptor mRNA Expression in Human Umbilical Vein Endothelial Cells after Lower Laminar Shear Stress

Abstract: Objective By using the technology of RT-PCR and Northern hybridization,to assess the expression of TLR-2 and TLR-4 in human umbilical vein endothelial cells(HUVECs) after lower laminar shear stress.Methods Total RNA was isolated from HUVECs untreated and treated with 42 μN/cm2 for 1 h.The technology of RT-PCR and Northern hybridization were used to determine the change of TLR-2 and TLR-4 mRNAs expression after 1 hour stimulation of laminar shear stress.Results The results of RT-PCR and Northern hybridization showed that the expression of TLR-4 in HUVECs was increased after 1 h shear stress.There was no change for TLR-2 after stimulation.Conclusion Lower laminar shear stress can induce TLR-4 mRNA expression.The experiment suggests that the inflammatory signal receptor TLR-4 would probably be involved in flow shear stress-induced effective gene expression in human vascular endothelial cells.
