Screening the Various Expression of Apoptosis Related Genes between Glioblastoma and Normal Brain Tissue with cDNA Microarray

Abstract: Objective To Screen the various expression of apoptosis related genes between glioblastomas and normal brain tissue with cDNA microarray.Methods Totally 12 tumor samples and 8 normal brain tissues were used in the experiments. Tumor samples were obtained from the patients who underwent operation in our department, and the control brain tissue were obtained from the patients of epilepsy who underwent lobe-ectomy in our department. The various expression of apoptosis associated genes between glioblastoma and normal brain tissue were screened by means of a cDNA microarray. The hybridized signal was normalized using the signal of a set of house-keeping genes. The gene whose signal change was more than 1.5 times were selected and considered at significantly different levels.Results Totally, 28 differential expression genes were found in the experiments including 17 up-regulation and 11 down-regulation. 6 of them were selected and corroborated using RT-PCR. The results of RT-PCR of the selected genes showed that the direction of expression alteration were identical with the results of cDNA analysis.Conclusion A lot of apoptosis related genes involve in the development of glioblastoma, and CRAF1, hHR23B are the new genes which related to glioblastoma.
