Expression and corticosteroid inhibition of arachidonate 15-lipoxygenase in chronic rhinosinusitis with nasal polyps

Abstract: Objective To detect the expression of arachidonate 15-lipoxygenase (ALOX15) in eosinophilic chronic rhinosinusitis with nasal polyps (ECRSwNP), and nonesinophilc chronic rhinosinusitis with nasal polyps (nonECRSwNP). Moreover, the effect of corticosteroid on ALOX15 expression was evaluated. Methods Samples were socked in RNA later and embedded in paraffin were acquired from library of Beijing Tongren Hospital. Real-time PCR and immunohistochemistry (IHC) were applied for mRNA quantification and protein distribution of ALOX15 in ECRSwNP, nonECRSwNP and healthy control group respectively.ALOX15 mRNA level of ECRSwNP samples pre-and post-corticosteroid treatment were detected. Results LOX15 mRNA level and protein expression were found increased significantly in ECRSwNP compared with nonECRSwNP and healthy control (both P=0.000). There was no significant difference between nonECRSwNP and healthy control (P=0.594, P=0.550). ALOX15 protein localized in a majority of cells including epithelia, plasma cells,neutrophils and eosinophils.One-week oral corticosteroid could decrease ALOX15 mRNA and protein expression in ECRSwNP significantly (P=0.004, P=0.024). Conclusion ALOX15 is upregulated in ECRSwNP compared with nonECRSwNP and control groups. Corticosteroid treatment could inhibit ALOX15 expression in ECRSwNP.
