Effects of iNOSmRNA on Pancreatic Endocrine Function of Experimental Severe Acute Pancreatitis in Rats

Abstract:

Objective To investigate the endocrine function changes in rats with severe acute pancreatitis, and explore the effects of iNOS on pancreatic endocrine function of experimental severe acute pancreatitis in rats.Methods SAP model was produced and the pathological score and the level of serum glucose and lipases were determined. Islets of rats with SAP were isolated and the amount of insulin secretion stimulated by glucose was determined by radioimmunoassay (RIA). The iNOSmRNA expression in isolated islet cells were assayed by reverse transcription PCR.Results In SAP models, pancreas showed pathological injury after induction of pancreatitis, accompanied with the elevated levels of serum glucose and lipases. The amount of insulin secretion induced by glucose from rats' islets with SAP were less than those from normal rats. In SAP rats, the iNOSmRNA expression of islet increased significantly.Conclusion SAP could impair the pancreatic endocrine function, and iNOSmRNA has been involved in the islet functional injure in SAP.
