    Objective To investigate the regulatory role of miR-365 on proliferation and tumorigenicity of gastric cancer cells, and explore the functional mechanism of miR-365 by verifying its target molecule E2F2 protein. Methods The expression profile of miR-365 in mouse tissues was checked by Northern blotting. The expression of miR-365 in human gastric cancer tissues was detected by Real-Time PCR. The regulatory role of miR-365 on gastric cancer cell proliferation and tumorigenicity were explored with cells experiment and nude mouse tumorigenicity assay; The potential binding site of miR-365 in the E2F2 3' untranslated region (3'UTR) was predicted with the bioinformatic tools; The luciferase report plasmids containing the wild type and mutated binding site of E2F2 3'UTR were constructed, and were used to study the regulation mechanism and identify the binding sites of miR-365 by luciferase activity analysis; The regulation effect of miR-365 on E2F2 protein expression was checked by Western blotting. Results The specific expression of miR-365 was detected in various digestive tissues including stomach; miR-365 was down-regulated in human gastric cancer tissues; miR-365 inhibited the expression of E2F2 protein by recognizing the specific binding site on the 3'UTR of E2F2 mRNA. Conclusion miR-365 was down-regulated in gastric cancer tissues of human and mouse model, and suppressed the proliferation and tumorigenicity of gastric cancer cells by inhibiting the expression of E2F2.
    Effect of DBHS family members on migration and invasion of esophageal squamous cell carcinoma

    Objective To investigate the effects of drosophila behavior human splicing(DBHS) family members, including p54nrb, PSF and PSPC1, on regulation of migration and invasion in human esophageal squamous cell carcinoma (ESCC) cell line TE-8.Methods After transfection with p54nrb, PSF and PSPC1 plasmids, the expression of mRNA and protein in human ESCC cell line TE-8 were determined by qRT-PCR and Western blotting, respectively. Wound healing and Matrigel invasion test were used to detect the effects of p54nrb, PSF and PSPC1 overexpression on migration and invasion of the cells, respectively. Results After 48 h of transfection with p54nrb, PSF and PSPC1 plasmids, the expression of relatives p54nrb, PSF and PSPC1 mRNA and protein levels were increased (P<0.05), respectiveIy. The migration rate was measured by wound healing after overexpression of p54nrb, PSF and PSPC1, was promoted significantly (P<0.05). Matrigel invasion of the cells were significantly increased after p54nrb, PSF and PSPC1 plasmids transfection (P<0.05). Conclusion Overexpression of DBHS family members could promote both migration and invasion of ESCC cell line.
    Effect of PHF8 gene on the growth of transplanted tumor of esophageal squamous-cell carcinoma in nude mice

    Objective To establish nude mice models with implanted tumor of esophageal squamous-cell carcinoma cells, and to observe the effects of PHF8 gene on the growth of implanted tumor. Methods Nude mice were injected subcutaneously into their right posterior flank with human TE-1 esophageal squamous-cell carcinoma cells (control group), lentivirus mediated Non-silencing shRNA TE-1 cells (Non-silencing shRNA group), or lentivirus mediated PHF8 shRNA TE-1 cells (PHF8 shRNA group), respectively. The size of implanted tumor was measured at regular intervals and the growth curve of tumor was portrayed. Animals were sacrificed at 4 weeks, and the silencing of PHF8 in tumor tissues was confirmed by real-time PCR and Western blotting.Results The tumorigenic effect on nude mice was weaker and lower in PHF8 shRNA group, as compared to those in Non-silencing shRNA group and control group. The expression of PHF8 mRNA and protein in PHF8 shRNA group was significantly decreased. Conclusion Lentivirus mediated shRNA effectively inhibited the expression of PHF8 in transplanted tumor of nude mice with human esophageal squamous-cell carcinoma cell lines, and the tumorigenic ability was significantly reduced.
    Association of DEC1 and cellular senescence of esophageal squamous cell carcinoma induced by cisplatin

    Objective To investigate whether cisplatin could induce senescence in esophageal squamous cell carcinoma (ESCC).Methods Four ESCC cell lines (ECA109, EC9706, TE-1, TE-3) and one normal esophageal epithelial cell line (Het-1a) were analyzed for this study. Reverse-transcription polymerase chain reaction (RT-PCR) and Western blotting were employed to detect the mRNA and protein expression of p53 and differentiated embryo-chondrocyte-expressed gene1 (DEC1) in ESCC cell lines. MTT and senescence associated β-galactosidase were used to explore an appropriate cisplatin concentration to induce proliferation inhibition and cellular senescence in TE-3 cell line. Further, Western blotting was employed to detect the expression of DEC1 and p53.Results In this study, we identified that p53 had a lower expression compared with normal esophageal epithelial cells by testing mRNA and protein expression of different esophageal cancer cell lines, while the expression of DEC1 had a higher expression. MTT assay showed that cisplatin inhibited the growth of TE-3 in a dose-and time-dependent manner. Combined MTT with senescence associated β-galactosidase, 4uM and 48 hours were chosen as most appropriate condition to induce cellular senescence. Elevated levels of senescence associated β-galactosidase were observed in TE-3 cells when exposed to low doses of cisplatin. TE3 cell experienced morphological changes following drug exposure accompanied with up regulation of DEC1 and p53.Conclusion Our results revealed that cisplatin could induce cellular senescence in esophageal squamous cell carcinoma. Meanwhile, p53 and DEC1 may be involved in this process.

    Objective To demonstrate drug resistance induced by serum starvation in gastric cancer and explore the possible mechanism.Methods The viability of SGC7901 cells was measured by MTT assay. The inhibition rates and IC50 values were then calculated. The phosphorylation level of EGFR and ERK induced by serum starvation were measured by Western blotting. Results Serum starvation could reduce the action of chemotherapeutic drug. EGFR signal activation acted as a protective factor against starvation by regulating downstream genes. Compared with the control group, the phosphorylation levels of EGFR and ERK were significantly increased in serum starvation treatment group (P<0.05). EGFR monoclonal antibody could partially reverse multiple drug resistance as a result of serum starvation. Conclusion Serum starvation may induce drug resistance in gastric cancer cell and the mechanism may be related to the activation of EGFR/ERK signaling pathway.

    Objective To investigate the risk factors for the nonalcoholic fatty liver disease(NAFLD) patients in railway staffs. Methods We measured the waist, height, and detected the serum fasting blood glucose, blood lipids, liver function, renal function and blood routine.Serum interleukin-6(IL-6), tumor necrosis factor-α(TNF-α) were detected using the method of liquid phase chip. Result Logistic regression analysis showed that the incidence and the seriousness of NAFLD were associated with higher body mass index(BMI), triglyceride, waist circumference and blood glucose. Serum IL-6, TNF-α were higher in patients with different severity of nonalcoholic fatty liver disease, especially in family members of patients iwth NAFLD. Conclusion The seriousness of NAFLD was associated with the level of metabolic disorders.IL-6 and TNF-α were much higher in family members of patients with NAFLD.

    Objective To investigate the feasibility of colorectal cancer screening being combined with the annual physical examination in a corporation. Methods A total of 5 270 people from a corporation in Yungang who had the annual physical examination in No. 731 Hospital of China Aerospace Science & Industry Corporation, aged from 40 to 74 years, were included in this study from April 2013 to July 2013. They completed questionnaires about the risk factor and accepted FOBT. Then those at risk and tested positive would have colonoscopy.Results All people completed the questionnaires and 62.87% of them received faecal occult blood test (FOBT). The rate of colonoscopy receiving was 30.33%. 22.28% of them who had colonoscopy were suffering from adenomatous polyp, and 24.75% of them were suffering from non-adenomatous polyp. Conclusion The receiving rate of colorectal cancer screening being combined with the annual physical examination is high and it can find colorectal precancerous lesions effectively.

    Objective To test the effect of sRAGE on myocardial apoptosis and STAT3 protein expression with or without STAT3 inhibitor AG490 following ischemia/reperfusion in vivo and in vitro. Methods C57BL/6J mice undergone left anterior descending coronary artery ligation were used as in vivo model and neonatal rat cardiomyocyte subjected to ischemic buffer as an in vitro model. Apoptosis was detected by TUNEL staining and caspase-3 activity. Expression of STAT3/p-STAT3 protein were detected by Western blotting analysis in the presence and absence of the JAK2 inhibitor AG 490. Results In vivo, compared with sham group, the number of TUNEL positive cells and caspase-3 activity were increased by 115% and 120%, and the ratio of p-STAT3/STAT3 was reduced by 50%; sRAGE (100 μg/day) reduced the TUNEL-positive myocytes by 51%, and activity of caspase-3 by 36%, increased the ratio of p-STAT3/STAT3 by 381% followed by I/R. In vitro, compared with control group, the number of TUNEL positive cells and caspase-3 activity increased by 380% and 77%, and the ratio of p-STAT3/STAT3 was reduced by 69%, sRAGE (900 ng/mL) reduced the TUNEL-positive myocytes by 63%, and caspase-3 activity by 33%, increased the ratio of p-STAT3/STAT3 by 243% followed by I/R. The effect of sRAGE reduction on TUNEL-positive myocytes and caspase-3 activity, raise of the ratio of p-STAT3/STAT3 were attenuated by STAT3 inhibitor AG490. Conclusion These results suggest that sRAGE protects cardiomyocytes from apoptosis induced by I/R in vitro and in vivo by activating STAT3.

    Objective To evaluate effective transcatheter atrial septal abnormalities (patent foramen ovale, PFO or atrial septal defect, ASD) closure rate and outcome in patients with ischemic stroke. Methods Ischemic stroke patients who underwent transcatheter closure of atrial septal abnormalities at Beijing Tiantan Hospital between May 2015 and November 2015 were enrolled. Right-left shunts (RLS) were confirmed by contrast transcranial Doppler ultrasound, and PFO or ASD were detected by transesophageal echocardiograph before transcatheter closure. Success rate of device implantation, completed closure rate and complications were evaluated. Results Ten patients [(48±14) years, 8 male and 2 female, 8 PFO and 2 ASD] were included in the study. Before closure, severe RLSs were detected in all patients. PFO size was (1.7±0.3) mm and ASD size was 4.5 mm and 3.4 mm. The procedure of device placement was successful in all patients. The 18/25 mm of PFO occluders were implanted in 8 PFO patients. The 12 mm and 10 mm ASD occluders were implanted in 2 ASD patients. Moderate residual RLS was present in 1 patient after PFO closure, and thus completed closure rate was 90%. Neither occluder malposition and thrombus,nor neurological recurrences were observed at 1, 3, and 6 months. Conclusion Transcatheter PFO or ASD closure is an effective and safe procedure in patients with ischemic stroke and can decrease RLS. However, it is necessary to evaluate residual RLS in mid-and long-term follow-up.

    Objective To report the experience of using a steerable sheath (Agilis NxT sheath) during the pulmonary veins isolation for paroxysmal atrial fibrillation (AF) ablation.Methods Twenty-six patients with paroxysmal AF (PaAF,n=26) underwent mapping and ablation using Agilis NxT sheath.The procedures of left atrium geometry was created undergoing mapping by EnSite VelocityTM v 3.0 after puncture of atrium septum and pulmonary veins angiography. Circumferential pulmonary veins isolation (CPVI) was performed using Agilis NxT sheath.Results Four PVIs were successfully performed in all 26 patients. The average operation time was (165.7±48.1) minutes, the average time of radiation was (9.4±0.5) seconds. The time of left atrial geometry modeling was (7.5±4.3) minutes, the time for left pulmonary vein isolation was (55±17) minutes, and the time for right pulmonary vein isolation was (37±11) minutes, respectively.Cardiac tamponade occurred in 1 cases, which was cured after pericardial puncture. Follow-up for 6-12 months showed that in 3 cases (11.5%) the AF recurred. Conclusion A steerable sheath is effective and safe during CPVI for AF ablation.
    Relationship between the lesions of coronary artery and the upper-aortic arteries stenosis

    Objective To explore the relationship between atherosclerotic heart disease and cerebrovascular disease through cardiac and cerebral angiography, to analyze the distribution characteristics of intracranial and extracranial sections lesions of upper-aortic arteries in patients with coronary artery disease (CAD). Methods According to the result of the coronary angiography (CAG),180 patients who were suspected to have coronary heart disease were divided into non-CAD group and CAD group. Cerebral angiography was performed in all patients, and all patients also provided information about physical history,age,sex,body mass index(BMI),smoking,and the results of blood routine tests,glycosylated hemoglobin,coagulation function,high sensitivity C-reactive protein(hs-CRP), homocysteine(HCY) and biochemical test were analyzed and the relationship between the lesions of coronary artery lesions and intracranial, extracranial sections of upper-aortic artery was explored,and the correlation of the two groups of subjects was analyzed with logistic regression model. Results There was no significant difference between CAD and non-CAD group in the intracranial carotid, extracranial carotid and intracranial vertebra(P=0.166,0.146,0.128). The incidence of extracranial vertebral artery stenosis in CAD group were significantly higher than that of non-CAD group(P=0.010).Conclusion Coronary artery lesions may be closely related to extracranial vertebral artery lesions.
    Relationship between circadian blood pressure rhythms and outcome in diabetic and nondiabetic patients with ischemic stroke

    Objective To investigate the association between circadian blood pressure rhythms and cardiovascular outcome in diabetic and nondiabetic patients with ischemic stroke. Methods The 373 ischemic stroke inpatients with large artery atherosclerosis were recruited and followed up at Cerebrovascular Center of Beijing Tiantan Hospital from May 2010 to August 2011. The baseline data were recorded. Ambulatory blood pressure monitoring was performed. The endpoint events included stroke recurrence, vascular events, and cardiovascular death. Results Totally 367 patients were included in the final analysis. The patients were divided into two groups according to the circadian blood pressure pattern: dipper (76 patients) and nondipper group (291 patients). The patients were followed-up for (19.0±5.0) months. The patients in nondipper group showed a higher prevalence of cardiovascular events, higher nocturnal blood pressure (P<0.05). Kaplan-Meier analysis showed that the cardiovascular events-free survival was lower in nondipper group than those in dipper group but did not reach statistical significance (P=0.052). Kaplan-Meier analysis showed that there were no significant differences in cardiovascular events-free survival between dipper and nondipper groups in nondiabetic patients (P=0.763); However, diabetic patients with nondipper showed a less cardiovascular events-free survival (P=0.030). Through Cox regression we found that the age (hazard ratio=1.037, 95%CI: 1.008-1.067, P=0.013), diabetes mellitus (hazard ratio=2.406, 95%CI: 1.340-4.319, P=0.003), modified. Rankin Scale (mRS) on admission (hazard ratio=1.379, 95%CI: 1.143-1.664, P=0.001) and nondipper (hazard ratio=0.964, 95%CI: 0.928-1.001, P=0.056) were associated with cardiovascular outcomes in diabetic patients with stroke. Conclusion Abnormal circadian blood pressure rhythms were associated with cardiovascular outcomes in stroke patients with diabetes, but we did not find a correlation between blood pressure rhythms and cardiovascular outcomes in stroke patients without diabetes.
    Study of PINK1 rescuing the mitochondrial dysfunction induced by α-synuclein

    Objective To identify whether PINK1 could rescue the mitochondrial dysfunction induced by α-synuclein. Methods MN9D cells were transfected with plasmid encoding human α-synuclein together with human wild type PINK1. The level of reactive oxygen species (ROS), mitochondrial permeability pore (mPTP) and mitochondrial membrane potential (ΔΨm) were examined by flow cytometry. The cell viability was observed by MTT assay and release of lactate dehydrogenase(LDH) respectively. Results While overexpression PINK1 could ease the opening of mPTP, reducing the generation of ROS, ΔΨm reduction percentage reduction by α-syn, reversed α-syn decreased cell viability and induced LDH release by α-syn induced. Conclusion PINK1 could alleviate α-syn-induced mitochondrial injury.

    Objective To investigate the effect of activation of CB1 on J774A.1 migration and to know whether HuR is involved in the process. Methods Immunofluorescence and agarose gel electrophoresis were used to detect the expression of cannabinoid receptor 1 (CB1) and human antigen R (HuR). Boyden chamber was used for cell migration assay. HuR siRNA transfection was used to determine whether ACEA(CB1 agonist) induced-migration depended on HuR. In order to observe its underlying mechanisms, HuR subcellular localization was assayed by Western blotting of cellular fractions from J774A.1 stimulated with ACEA.The effects of HuR siRNA on CB1 mRNA and protein levels in J774A.1 stimulated with ACEA was assayed by Western blotting. Results In this study, we present evidence that J774A.1 significantly expressed HuR and CB1; the treatment of ACEA caused a increase in J774A.1 migration(P<0.01), and pharmacological inhibition of CB1 markedly attenuated ACEA-induced migration; ACEA-induced J774A.1 migration depended on HuR. Furthermore, we demonstrated ACEA increased HuR cytoplasmic translocation, and HuR regulated the expression of CB1. Conclusion The activation of CB1 induces J774A.1 migration and HuR is involved in the process.

    Objective To explore the research situation in abroad of liver fibrosis in chronic liver disease the last ten years. Methods The English literatures which were involved in liver fibrosis diagnosis from 1995 to 2014 have been retrieved on PubMed, Embase, Web of science. The bibliometric analysis has been operated with Endnote X7, Bicomb2.0 software.The high frequency keywords of liver fibrosis diagnostic test have been defined by the frequency not less than 40. At last, the high frequency keywords have been put into SPSS20.0 and made cluster analysis.Results A total of 1 253 available literatures have been screened, roughly the amount of literatures published on the rise. The 5 767 key words of liver fibrosis diagnostic test have been screened. The most frequent key words are biopsy,biological markers and elastography in the order. Conclusion Biopsy is still the diagnostic reference standard of liver fibrosis. Serological examination is common testing method. Imaging,especially ultrasonic elasticity measurement has been major noninvasive diagnostic means.
    Effect of hypoxic preconditioning on the serum levels of TNF-α,IL-1β and IL-6 in mice with acute cerebral infarction

    Objective To investigate the effect of hypoxic preconditioning on the serum levels of tumor necrosis factor alpha(TNF-α), interleukin-1 beta(IL-1β) and interleukin-6 (IL-6) in rats with acute cerebral infarction.Methods Twenty-four rats were randomly divided into the four groups: normal control group(normal,N), sham operation group(control,C), acute cerebral infarction group(cerebral infarction,CI), hypoxic preconditioning(hypoxic preconditioning,HP) and acute cerebral infarction group(HP+CI), with 6 rats in each group. Models of hypoxic preconditioning and photochemical acute cerebral infarction were duplicated.Behavioral method and ELISA were used to respectively measure neurological deficit scores and the levels of TNF-α, IL-1β and IL-6.Results No symptom of neurological deficit is shown in group N and C,while it showed up in group CI and HP+CI. The neurological scores in group CI and HP+CI decreased more clearly than those in group N and C (P<0.05). The neurological scores in group HP+CI improved significantly than those in group CI (P<0.05). The serum levels of TNF-α, IL-1β and IL-6 in group CI and HP+CI increased significantly than those in group N and C (P<0.05). The serum levels of TNF-α, IL-1β and IL-6 in group HP+CI decreased more clearly than those in group CI (P<0.05).Conclusion Hypoxic preconditioning, can protect animals from acute cerebral infarction and the protection may related to inhibition of inflammatory reaction (subsequent to acute cerebral infarction) induced by the decrease in release of TNF-α, IL-1β and IL-6.
    Clinical and pathological analysis of 7 cases of primary cardiac tumors in children

    Objective To investigate the histologic types and clinical features of primary cardiac tumors in children. Methods After observation of the gross specimen, we got the tissue sampling along the tumor's maximum section, and did the conventional paraffin section, HE staining,histochemistry and immunohistochemical staining, microscopic observation, and analysed the clinical data. Results According the morphological characteristics and the results of immunohistochemistry, 7 cases were all benign, including 3 cases of rhabdomyoma, 3 cases of cardiac fibroma, 1 case of papillary fibroelastoma. Conclusion Primary cardiac tumors in children were rare.Most of them were benign.Mark of immunohistochemical and special histochemical staining could help the diagnosis.It was very important for treatment and prognosis of primary cardiac tumors to make accurate diagnosis,and effective surgical procedures.
    Radiation effective dose received by performing CT-guided interventional procedures in the patients with liver cancer

    Objective Totally To explore the effective radiation dose taken during 5 kinds CT-guided interventional procedures to the patients with liver cancer. Methods Totally 390 case-time inspection records from Sept. to Dec. 2014, of who were diagnosed and treated by the CT-guided interventional procedures, were retrospectively analyzed. The interventional procedures include liver biopsy, chemical ablation, radiofrequency ablation, microwave ablation, and cryoablation. The type of CT-guided interventional procedure, the scanning time, the exposure dose(total mAs),volume-weighted CT dose index(CTDIvol) and the total does-length product (DLP) were recorded. Effective dose(ED) was calculated. Results The average scanning time of liver biopsy, chemical ablation, radiofrequency ablation, microwave ablation, and cryoablation were (49.7±23.4)s,(88.9±21.3)s,(89.5±40.6)s,(106.0±43.5)s and (87.7±27.6)s. The EDs of the five procedures were (19.5±14.9) mSv,(30.0±10.2) mSv,(34.2±17.6) mSv,(38.1±19.9) mSv and (33.1±13.1) mSv, respectively. The EDs in biopsy procedure were significantly lower than those in ablation procedures. It was represented that the ED are positively correlative with the tube current, the scanning time, the exposure dose and the CTDIvol. Conclusion The effective radiation dose of single CT-guided interventional procedure is relatively lower. Therefore the risk of radiational damage and subsequent complications is lower as well. It should be paid attention that the cumulated effective dose for several times of CT-guided interventional treatments may exceed the acceptable range.
