Measurement of Histamine in Nasal Mucosa in Guinea Pig with Allergic Rhinitis

Abstract: Objective To observe the content of histamine in nasal mucosa in allergic rhinitis(AR) guinea pig,and study the role of it in the diagnose of AR.Methods In the Shore fluorescent method,two factors: temperature and extraction time that affected the measurement of the histamine were estimated respectively at two levels by the recovery rate of histamine to build the final method of measurement of histamine.In addition,the allergic rhinitis models in guinea pig were built by using sensitizing agent TDI,and the histamine in nasal mucosa in guinea pigs were measured by the method built above.Results At the condition of 25 ℃ and 45 min of extraction time,the recovery rate of histamine measured up the requirement.And by the method,the histamine content in nasal mucosa in controlled group and AR group were(513.7±49.3)ng/g and(590.4±47.6)ng/g respectively.There were significant difference between natural controlled group and AR group(P<0.01).Conclusion The improved Shore fluorescent method could accurately and reliably measure the histamine in nasal mucosa in guinea pig.The histamine content in nasal mucosa played an important role in diagnosing AR disease.
