The objective was to study the effects of brain-derived neurotrophic factor(BDNF)and neurotrophin-3(NT-3)on the growth and development of cultured spinal cord neurons of fetal rats. The survival and growth condition of cultured spinal cord neurons were observed under phase-contract microscope. The cell numbers were counted. The body diameter and process were measured. All data obtained were expressed by
x
±s and analyzed by variance analysis and SNK test. Both BDNF and NT-3 can increase survival rate of neurons in vitro. Effect of BDNF is more significant than NT-3. BDNF can enhance the development of cell bodies and NT-3 can promote the outgrowth of processes. BDNF and NT-3 have specific effects on growth and development of cultured spinal cord neurons.

A simple and sensitive HPLC method for the determination of drotaverine in human plasma has been developed. Drotaverine in alkalinized plasma was extracted with aether solvent and the drotaverine in the aether phase was back-extracted into 0.1 mol/L HCl. 10μL of the aqueous layer was injected into the C18-column and the eluent was monitored at 254 nm. Separation was performed on the column with 0.02 mol/L sodium dihydrogen phosphate-methanol(30∶70)containing perchlorate at pH 2.8 as mobile phase. Drotaverine was well resolved from the plasma constituents and internal standard. An excellent linearity was observed between peak-height ratios and plasma concentrations and the RSD was always <15%. The lowest limit of detection(signal-to-noise ratio 3∶1)was 8 μg/L, the recovery rate was between 80%~100%. The method is suitable for therapeutic monitoring and pharmacokinetic studies of drotaverine in humans.

Objective: To investigate the pathological course in the intracranial aneurysms. Methods: One case normal intracranial artery(from cortex fistulization), 11 cases ruptured aneurysms, two cases unruptured aneurysms were obtained from neurosurgical excision. Routine HE stain was used to observe histological characteristics, immunohistochemistry was used to observe the expression of monocyte chemoattractant protein-1(MCP-1)and macrocyte inflammation protein-1α(MIP-1α)in the walls of the normal artery and aneurysms(unruptured and ruptured). Result: By the HE strain the wall of the ruptured aneurysms(10 cases)and unruptured ones(2 cases)consisted of incrassated intima and connectivum extima. The fibroblast in the intima was arrayed in the disorder. Monocyte-like cells could be seen in the whole aneurysm wall. In one case of aneurysm wall(ruptured)glass-like fiber structure was left over, and few cells could be seen. Nine cases of mural thrombus could be found. The thrombus represented with organization. Immunohistochemistry: MCP-1 and MIP-1α were not detectable in the normal artery; the immunohistochemistry signal of MCP-1 and MIP-1α could be observed upregulated in the ruptured aneurysms(10 cases)and unruptured ones(2 cases), often in the intima. MCP-1 and MIP-1α appeared to be expressed by Fibroblast cell in its cytoplasm. Monocyte-like cells had little cytoplasm, the signal was seldom seen. The immunohistochemistry signal was discontinuous in the intima, MCP-1 and MIP-1α expressed where fibroblast and monocyte-like cells assembled. One case of ruptured aneurysms had no signal because there were no cells, only glass-like fiber. Mural thrombus had upregulated signal of MCP-1 and MIP-1α in the cytoplasm of fibroblasts and endotheliocytes of its micrangium. Conclusion: the pathological representation of the ruptured and unruptured aneurysms and the upregulated expression of MCP-1 and MIP-α in the aneurysm wall suggest that the development of the aneurysm may be a course of the chronic inflammation whose main inflammatory cells are monocyte-like cells.

Although cyclosporine and antibiotics have been widely used in clinical for immune suppressing in organ transplantation and antimicrobial in bacterial infection respectively, but their effects on CD4
+
and CD8
+
lymphocytes infiltration in myocardium have been unknown. In the present study we accounted CD4
+
and CD8
+
lymphocytes by immuno-histochemistry, in myocardium of rats which had separately orally administrated with cyclosporine and antibiotic and selenium for 24 h, and compared to the control group. Results showed that both of CD4
+
and CD8
+
lymphocytes accounts were significantly decreased(
P
<0.01)in three experimental groups than the control. The cell accountings towards cyclosporine were lower than antibiotics and lower than selenium. These data suggest that all of cyclosporine and antibiotics and selenium evidently suppress CD4
+
and CD8
+
lymphocytes infiltration in myocardium, and display a different suppressing.

To determine the clinical efficacy of amiodarone in fast atrial fibrillation in patients with acute myocardial infarction, 48 acute myocardial infarction patients with recent onset of fast atrial fibrillation were observed. After an intravenous load, intravenous amiodarone was administered continuously. Efficacy of amiodarone in the reversion of atrial fibrillation and the control of ventricular rate as well as the side effects were observed. The ventricular rates of 15 min, 30 min, 1 h, 6 h, 24 h after the administration of amiodarone in the 48 patients were(107.17±9.67)beats/min,(96.00± 8.39)beats/min,(88.13±9.98)beats/min,(79.88±10.19)beats/min,(74.88±9.80)beats/min respectively, which were lower than the rate(130.58±9.15)beats/min before the drug was given. Atrial fibrillation was reverted to sinus rhythm within 24 h in 48 cases. Sinus bradycardia was observecd in 3 patients. Sinus bradycardia and Ⅱ°Ⅱ type AVB occurred in 1 patient. Intravenous amiodarone is effective and safe in the treatment of fast atrial fibrillation in patients with AMI.

To study the relations between arrhythmia and left ventricle hypertrophy(LVH)in patients with essential hypertension, we examined 90 patients with essential hypertension by 24 h dynamic electro-cardiogram and echocardiogram, and analyzed the relations between arrhythmia and LVH and myocardium ischemia. Out of the 90 patients, 47(52.2%)were found suffering ventricular arrhythmia and 61(67.8%)were found suffering atrial arrhythmia. The incidence rate of atrial arrhythmia was higher than ventricular arrhythmia. The incidence rate of ventricular arrhythmia among patients with LVH was evidently higher than that among patients without LVH. The incidence rate of ventricular arrhythmia among patients suffering hypertention with the change of ST-T was evidently higher than that among patients suffering hypertention without the change of ST-T. The incidence rate of hypertensive arrhythmia is close related to LVH.

The objective was to evaluate the long-term effect of α-interferon(IFN)thrapy in patients with chronic hepatitis B virus infection, particularly on survival and hepatocellular carcinoma(HCC)prevention. Fifty-seven cases were treated with interferon(3~5MIU), 42 cases were treated with general therapy in contrast group. They were followed up for 2.3~5.5 years after end of therapy. Follow-up studies included clinical, biochemical and virological aspects and HCC screening every 3 to 6 months. In the treatment group, the HBeAg serum transforming negative and HBV DNA transforming negative were 40.4%、46.3%、42.1%、51.9% separately in the end of treatment and the end of follow-up, and was higher than the control group(
P
<0.01). The cumulative incidence of cirrhosis and HCC development was higher in the control group than in the treated group, In contrast, the cumulative survival rate was higher in the treated group. The results suggest that interferon has long-term beneficial effect in terms of HBV clearance, reduction of HCC and prolongation of survival.

To determine and evaluate the changes of free radicals in brain tissue after brain injury and the relationship between them and brain edema, 42 SD rats were used and divided into three groups: normal controls, sham group and operation group. The cerebral contusion model was produced by weight dropping. The brain injury model was built successfully. The contents of SOD and MDA in cerebral contusion tissue were measured, and so were brain water contents. The data from the normal control group and the sham group showed no significant difference. MDA values of the traumatic right hemisphere of the operation group were significant higher than that of the normal control group and sham group, so were brain water contents(
P
<0.05). SOD activity of the traumatic hemisphere was lower than that of the other two group(
P
<0.05). The results of this study suggests: ①lipid peroxidation takes place in the traumatic cerebrum, and free radicals play the role in tissue damage due to ischemia and bleeding; ②oxidative stress causes brain edema and vice versa.

The objective was to summarize the experience in the diagnosis and treatment of 85 cases of thoracic injury combined with head injury in order to improve the standard of diagnosis and treatment. We retrospectively reviewed 85 cases of thoracic injury combined with head injury. The main cause of thoracic injury combined with head injury was traffic accident(68 cases,80%). Operative treatments were employed in 14 cases. Eighteen cases(21.2%)died, of which 11 cases(61.1%)died of head injury, 3 cases(16.7%)of shock due to acute blood loss, 3 cases(16.7%)of ARDS, 1 case(5.5%)of MSOF. The results suggest that it's very important to realize the seriousness of head injury. The treatment should be performed with diagnosis at the same time. The injury history and physical examination are helpful to the diagnosis of head injury by injury history and physical examination. The dangerous and serious injury should take the priority of the treatment. It should be carefully dealt for the paradorical management between shock and intracranial hypertension. Attention should also be paid to the increase of the incidence and the mortality of ARDS in the thoracic injury combined with head injury.

Expression of bcl-2 and P53 proteins in twenty-eight cases of prostatic carcinoma and ten cases of benign hyperplasia of prostate was detected by immunohistochemical LSAB assay to study the relationship between these gene products and the pathogenesis of prostatic carcinoma. It was found that bcl-2 and p53 positive rates in prostatic carcinoma were 42.9% and 28.6% respectively, both of which were not significantly higher than those in benign hyperplasia of prostate statically, but a tendancy of their increasing expression in prostatic carcinoma could be seen. The expression of bcl-2 in the group of poor differentiation was significantly higher than that in the well-differentiated group(
P
<0.05). When compared in terms of clinical stages, the bcl-2 expression at stage D was significantly higher than that at stage B or C(
P
<0.05). There existed a tendency of increase in p53 expression in prostatic carcinoma, but it was not significantly higher than that in benign hyperplasia of prostate statistically. When the poorly differentiated group and well-differentiated group were compared, there was no statistically significant difference, and the comparison based on different clinical stages didn't show significant difference in p53 expression between stage and Stage B or C. It is suggested that bcl-2 protein expression increases in prostatic carcinoma and is, to some extent, involved in the pathogenesis of the disease. It may be concluded that bcl-2 protein assay can be used as an index of diagnosis and prognosis of the disease. The relationship between p53 expression and prostatic carcinoma remains to be learned.

The objective was to evaluate the relationship between the pathological features of umbilical vessels and placenta, the S/D ratio, and visual disturbances in pregnancy induced hypertension(PIH). The umbilical vessels and placenta of 41 women with PIH and 23 normal term-pregnant women(control group)were observed under light microscopy and a special staining method, doppler and ultrasound S/D ratio. The ophthalmologist examined the visual disturbances. Forty-one women with PIH were categorized into three groups according to the degree of lesions: no lesion(
n
=2), mild(
n
=17),and severe(
n
=22). The pathological changes of umbilical vessels and placenta in moderate and severe PIH were more severe than those in mild PIH(
P
<0.01). However, there were no apparent pathological changes in the control group. The pathological changes of umbilical vessels and placenta in PIH had a significant relationship with S/D ratio, and neonatal outcome. The pathological changes of umbilical vessels and placenta can reflect the characteristic lesion of PIH and are parallel with the severity of the disease. The pathological changes of umbilical vessels and placenta are parallel with the S/D ratio and the visual disturbances. The pathological changes of placenta are more severe than umbilical vessels.

In order to evaluate clinical features and levels of aspartate aminotranferase(AST)in gingival crevicular fluid of gingivitis in adolescents, 21 healthy and 28 gingivitis subjects, age range from 10 to 16, were selected. The clinical parameters were examined and recorded. Within each subject, the gingival crevicular fluid(GCF)sample was collected. The value of GCF and AST were measured by biochemical analysis instrument. The clinical parameters of gingivitis in adolescents were the plaque control record(PCR): 73%, oral hygine index(OHI): Ⅱ(36%), Ⅲ(64%); gingival index(GI):Ⅰ(25%), Ⅱ(43%), Ⅲ(32%)and gingival bleeding(GBI): 54%. The amounts of GCF, GCF-AST were significantly higher in samples from gingivitis group than in the healthy group. Gingivitis in adolescents have a risk of disease progress. AST in GCF may also be a useful marker for gingivitis tendency.
