To explore the molecular mechanism of hypoxic preconditioning, isolate and analysis the differential genes from the hippocampus of hypoxic preconditioned mice, the technology of mRNA differential display was used and 56 bands of differential genes were recovered. Fourteen of them were purified, cloned, and sequenced. The BLAST results show that nine of them are known genes, while five of them may be unknown genes. The differentially expressed genes will provide a new clue for further study of the molecular mechanisms of hypoxic preconditioning.

To investigate the effect of NGF on neurons in the dorsal root ganglion(DRG)after a injury of peripheral nerves, a unilateral transaction of the branchial plexus was done in six rabbits with the age of one month. After the transaction, 25 μg NGF was given to the stump end of the nerves before they were sutured and all animals were kept alive for one month. During this period, the rabbits were intramuscularly injected with 12.5 μg NGF every other day. The sizes of the cell bodies and nucleoli and numbers of neurons in the DRG were observed and counted under a light microscope. The ratio of the cell numbers between the injured side and uninjured side were quite close to one in the six rabbits. The nucleoli were taken as a index of calculation of cell numbers. The image analysis was used to measure the sizes of cells and nucleoli in one rabbit. The statistic analysis were used to make comparison of data from different segments and we found that there was no remarkable difference with regard to average diameter of the cells between the injured and uninjured side(

>0.05). There was noremarkable difference on the average diameter of the nucleoli between the two sides either(

The objective is to study the effects of 11,12-epoxyeicosatrienoic acid(11,12-EET)on the cardio pulmonary cerebral resuscitation(CPCR). Using the classical method to reproduce the resuscital model, we bring the CPCR into effect at the moment when the heartbeat stopped. Then we observed the succeed rate of resuscitation, the resume rate and time of independent circle and the change of the left heart function. Preconditioning with 11,12-EET or applying 11,12-EET in CPCR can enhance the success rate of CPCR, shorten the resume time of independent circle and improve the heart function after CPCR. The study indicates that 11,12-EET can improve the prognosis of CPCR.

The aim of this study is to investigate mechanisms of duodenal motility disorders presented in cold restraint stress rats by using electrode and highly sensitive sensor to record alterations of duodenum electromechanical activity. The study suggested that the average frequency and amplitude of duodenal slow wave was lower than the control group(

To investigate the effect of hyperbaric oxygen(HBO)on caspase-3 activity, we devided 200~250 g Sprague-Dawlay rats into three groups: the ischemia/ reperfusion group(I/R), the group treated by HBO and the sham operated group(Sham-O)as the control. Global brain ischemia was induced by using a modified four vessel occlusion model originally described by Pulsinelli and Buchan. Cortex of parietal lobe was obtained and homogenized respectively at reperfusion time points of 6 h, 24 h, 48 h and 96 h after 20 minutes of ischemia. Then caspase-3 activity was measured by cleavage of the colorimetric substrate of DEVD-ρNA. Although both caspase-3 activity of the HBO group and I/R group were not statistically different from the Sham-O group at reperfusion time point of 6 h,the HBO group was higher than the I/R group(
P
<0.05). Both the HBO group and I/R group were higher than the Sham-O group(
P
<0.05)at 24 h of reperfusion, but there was no statistical difference between the former two groups. At reperfusion time points of 48 h and 96 h, the HBO and I/R group were higher than Sham-O group, but there was no statistical difference. And there was no statistical difference between the former two groups either, which indicated that caspase-3 activity had decreased to normal levels. The results of above indicated that caspase-3 was activated after global ischemia/reperfusion. HBO treatment at early time may aggravate this activation, but after several times of HBO treatment, this promotion effect becomes indistinct.

TCSs, one kind of Chinese traditional herbs, is the serials of anticancer medicine. The purpose is to study the mechanism of tumor inhibitory and killing effects of the TCSs on liver cancer cell line 7721. With methods of flow cytometry and Western blotting, we investigated tumor cell apoptosis induced by the herb. It also examined the expression of apoptotic protein with cytochemistry method. The results indicated that the herb can induce tumor cell apoptosis, and the apoptotic rate can reach to 40 percent. After being treated by the herb, the expression of antiapoptosis gene bc1-2 reduced, and proapoptosis gene Bax increased. The results suggest that to induce target cells apoptosis is the main mechanism to inhibit and kill tumor by the herb.

The objective was to investigate if melatonin is related with the decrease of MDA by pinealectomy mice during long term aerobic exercise. Pinealectomied 12-month-old mice were divided into three groups: the control, the swimming-60-min and the 60-min with 3% body mass; and normal mice also were divided three groups in the same way. After eight weeks swimming, all animals were killed, and their MDA levels in serum were assayed. Results: In male mice MDA decreased significantly in normal mice than in the pinealectomied in the swimming-60-min and the swimming-60-min with 3% body mass. while in female mice, the swimming-60-min group has shown the same results as above, but in the swimming-60-min with 3% body weight group the level MDA in pinealectomied group decreased significantly than in the normal group. Conclusion: long term suitable intensity aerobic exercise can enhance the activity of antioxidase, and it is related with stimulating pineal and releasing melatonin.

Various methods were used to culture rat embryonic neural stem cells(NSCs). The cells were induced to divide so as to be confirmed as NSCs, and were checked by phase contrast microscopy and immunofluorescence. The results showed that cells grew the fastest when both bFGF and EGF were added into the culture medium. Higher NSCs concentration promoted cell proliferation. It suggests that the two neurotrophic factors have cooperative effects on stimulating cell growth and higher NSCs culture concentration can promote its proliferation.

P4501A1 is a kind of enzyme related with the carcinogenic activity of aromatic hydrocarbon, the polymorphism in exons 7 of the gene could alter the function, and has been reported to be associated with lung cancer in Japanese. But data from Caucasian or African-American were different. The difference is said to be from the different frequencies of this polymorphism between ethnic groups. This paper analyzed the frequency of polymorphism in exons 7 of CYP1A1 gene in Chinese by the PCR method. The results suggested that the frequencies of genotype in Chinese were Ile/Ile 61.6%, Ile/Val 26.4% and Val/Val 12.0%, no significant difference of the frequencies between male and female was found. But the frequency of Val/Val was much higher than that in Japanese, Caucasian or African American.

In order to evaluate the accuracy of left ventricular ejection fraction(EF)and fractional shortening(FS)for predicting malignant ventricular arrhythmia(VA)after coronary artery bypass graft(CABG). One hundred and fifty consecutive patients were selected for an isolated CABG procedure, EF and FS, left ventricular potential(LVP), myocardial enzymes and 24 h Holter were measured in special perioperative period. 1)Patients with previous myocardial infarction(MI), aneurysm, positive LVP had obviously lower value of EF and FS before CABG than those without them. 2)In patients with LVD, significant improvements were found in EF and FS indexes after CABG. 3)The incidence of postoperative VA is higher in these patients with LVD, VA, VLP+ and aneurysm than the others. The results showed that: 1)EF and FS were sensitive indexes in reflecting left ventricular function. 2)CABG and aneurysmectomy not only improved myocardial blood supply, but also increased the heart out put, and brought the best benefits to those patients with LVD. 3)For the reason of myocardial depression, the cardiac function of patients without LVD decreased in short term follow up(14±1)d. 4)EF≤40% and/or EF≤24% were independent predictors for the prevision of malignant VA, the value of FS accurately reflected left ventricular function. 5)Integrate with other indexes, such as LVD, VLP and aneurysm, would improve the sensitivity, specialty and accuracy for predicting malignant VA after CABG.

Fifty one uremic patients over 60 who received cadaveric renal transplantation were retrospectively analyzed in order to investigate the renal transplantation in the aged uremic patients. Results: the survival rate of person/kidney one year was 88.2%/86.2%. Two patients died from the acute tubular necrosis and heart failure and lung infection, one patient died from the severe pneumonia while the other one died from cerebrovascular accident. The total complications post transplantation included one hyperacute rejection, four acute rejection, two chronic rejection, four acute tubular necrosis, 13 hepatotoxicity and nephrotoxicity of cyclosporin A, six leukopenia, 2 posttransplant diabetes mellitus, 9 lung infection, eight heart failure and one fistula in the ureterocystanastomosis. Therefore, the advanced patients are still suitable for the renal transplantation. It special attention has to be paid on the treatment of infection and cardiovascular diseases and nursing care, and it is also important to adjust and monitor the toxic side effects of the immunosuppressive drugs, and the dose of CsA in the aged patients should be lower.

To study the lateral location diagnosis value of PET in temporal lobe epilepsy before operation, 20 patients with refractory temporal lobe epilepsy were investigated by EEG, MRI and PET. Nineteen of 20 patients had epileptiform discharges in EEG scalp recordings, but only one patient had normal EEG. EEG recordings showed unilateral EEG abnormal focus in 10 patients and bilateral abnormal focus in nine patients. MRI demonstrated 12 normal, five unilateral hippocampal atrophy, one left parietal occipital softening and one left frontal heterotopic gray matter and one rignt frontoparietal cyst accompanied with pituitary adenoma. PET revealed focal hypometabolism in all 20 patients, of whom 16 had unilateral abnormalities and 4 bilateral abnormalities. All patients were treated by operation, 16 patients underwent anterior temporal resection and hippocampectomy, 4 patients selective hippocampectomy. The pathologic examination showed gliosis and neuron death in resected samples. These results indicates that PET is superior to MRI in localizing diagnosis of temporal lobe epilepsy, more sensitive and reliable.

To study clinical and electrophysiological features in myotonic dystrophy. Methods: Electromyography(EMG)and nerve conduction velocity were performed in twenty-four patients with myotonic dystrophy. Results: In 46% of cases,(motor/or sensory)nerve conduction velocity was abnormal in at least two separate nerves. It was characterized by mildly slowing motor/or sensory nerve conduction and reducing amplitude of compound motor action potential(CMAP)/or sensory nerve action potential(SNAP). The severity of disease was associated with the patient's age. Conclusion: Peripheral neuropathy in myotonic dystrophy is not rare. It is a sensorimotor axonal peripheral neuropathy. The clinical symptoms are worse in older patients.

Induced abortion with propofol anaesthesia avoids patients from pain, it is a safe and effectual method to terminate pregnancy. However, propofol can not make cervix uteri relax, so the difficulties and complications from cervix uteri in induced abortion can not be resolved. In this study, the dosages and time of taking misoprostol orally before operation was studied further. It was proved that cervix uteri in experiment groups was much flabbier than that in control group, if women took misoprostol 200 μg or 400 μg orally 2 h before the operation. The effect was obvious. The method is worth popularizing.

The objective was to compare and investigate the interference and the difference of Creatinine's enzymatic method and kinetic Jaffe method. To measure creatinine of serum or plasma, Beckman CX 7 analyzer with Beckman reagent for kinetic Jaffe method and Randox reagent for enzymatic method were used. The interferences of lipemia, hemoglobin and bilirubin were compared. The results showed that enzymatic method was not interfered, Jaffe method was negatively affected by bilirubin(>158 μmol/L). Creatinine's enzymatic method is better than kinetic Jaffe method.
