    In order to obtain complete sequence and catalytic core of human tyrosine hydroxylase(TH) cDNA,the full length of cDNA from human TH was used as template and the specific oligonucleotide primers were designed for PCR.The target fragments were cloned into vector pGEX-4T-1 respectively.The recombinant plasmids were transferred into E.coli BL21,and the expression of target protein was induced with IPTG.The expressed protein was analysed by SDS-PAGE and western-blotting.The result showed that the cDNA fragments of complete sequence and catalytic core of human TH were harvested.The sequencing for them showed that the cloned target gene fragments were correct.SDS-PAGE indicated that the proteins with Mr about 83000 and 66000 were expressed with the recombinant plamids pGEXTHa and pGEXTHc respectively.

    Projection from the vestibular nuclei to the spinal cord was examined following injection of cholera toxin subunit B at various levels of the spinal cord in the rat. The results were as follows: ① The majority of the labeled cells were seen in the caudal ventral of LVN, caudal of DVN and MVmc of MVN from the injections into the upper cervical segment. ② The labeled cells were only seen in the caudal ventral of LVN, and distributed diffusely in the DVN and MVN from the injections into the cervical enlargement segment. ③ The labeled cells were distributed in all parts of LVN and DVN, especially densest in the rostral dorsal of LVN, and seldom seen in the MVmc of MVN from the injections into the thoracic and lumbar segment. ④ The labeled cells from the injections into the sacral segment distributed sparsely in the LVN and DVN. The result indicates that the fibers of origin from the DVN and LVN projected directly to the sacral segment, while the fibers of origin from the MVN only projected to the upper lumbar segment.

    The research is to study the influence on mouse skin graft survival with anti-CD3 antibody and donor bone marrow infusion. Under the effect of the immunosuppression of anti-CD3 antibody, bone marrow cell suspensions were extracted from donor strain C57BL/6 and injected into recipient BALB/C after skin allograft. The MST is 15.6 d for the bone marrow group, 10.1 d for the anti-CD3 group and 7.7 d in the contrast group respectively. The donor specific bone marrow infusion could improve the skin graft survival under the immunosuppression.

    The distribution of tau protein in the brain was studied in the groups of the healthy, controled mice, diabetic mice, and diabetic mice treated with APP17mer peptide, in order to examine the effect of APP17mer peptide on phosphorylation of tau protein and its potential to treat neurodegeneration. The diabetic mouse model was created with STZ. APP17mer peptide was used for treatment. Four weeks later, cryostat sections of the brain was prepared for immunohistochemical study with Tau 1 monoclonal antibody and after dephosphorylation with phosphatase PP-2B. On staining with Tau-1 the brain of the healthy mice and the diabetic mice treated with APP17mer peptide showed numerous immunopositive cells in the granular layer of the cortex, and in the hippocampus. The staining was dark (P<0.05). Only after dephosphorylation with PP-2B did the number and staining of immunopositive neurons in diabetic mice reach the level in healthy mice. The results demonstrate that tau protein is phosphorylated at Ser202/Ser205 in diabetic mice. APP17mer peptide can prevent the phosphorylation of tau protein at these sites.

    In order to evaluate combined serum tumor markers in the diagnosis of gastrointestinal malignant tumor, serum CA242 and CEA were concomitantly determined by ELISA in 95 patients with gastrointestinal malignant tumor, 90 patients with benign diseases and 30 healthy blood donors. In general, serum CA242 and CEA levels were increased more significantly in malignant tumor group than in the other two groups. The sensitivities of CA242 and CEA in gastrointestinal malignant tumor were 43.16% and 45.26% respectively, while the combined markers was 62.11%, which was significantly higher than either of the one marker(P<0.01). The specificity of the combined markers was 73.55% including colon tumors(72.41%), gastric carcinoma(69.23%), hepatic carcinoma (57.14%), pancreatic carcinoma (55.56%), which were all much higher than esophageal carcinoma(18.18%). These data suggest that the combined determination of serum CA242 and CEA can increase the sensitivity in diagnosis of gastrointestinal malignant tumors.

    From Jan 1996 to May 1999,the antimicro bial susceptibility of Escherichia coli and Klebsiella pnenumoniae to ceftazidime(CAZ), aztreonam(AZT), cefotaxime(CTX), ceftriaxone(CRO) and imipenem(IMP) were tested.The minium inhibitory concentration(MIC) testing from Jan 1996 to May 1999 was made for the antimicrobial suscepitibility of E.coli and K. pneumoniae; the Disk diffusion from May 1999 to Oct 1999 was tested for ESBLs in E.coli and K.pneumoniae.The results showed: ① the susceptibility in AZT increased; ② the susceptibility in CAZ, IMP was still strong; ③ the susceptibility of K.pneumoniae to CAZ, CTX, CRO in 1999 decreased obviously; ④ through the method of K-B, it was found that the rate of ESBLs pro duction in E. coli and K. pneumoniae in 1999 is 23.1%(21/88), the rate of K.pneumoniae in these 21 ESBLs-producing becteria is 61.9%(13/21), which is much higher than that of E.coli(38.1%, 8/21).

    To evaluate the special stain ing effect of potassium ferricyanide on vessel basement membrane, using the K3Fe(CN)6 to treat the biopse specimen and examine the reaction of basement membrane to it under the electron microscope. The result showed the potassium ferricyanide increased the electroncontrast of vessel basement membrane and clearly showed the ultrastructure of it. The staining method provides a kind of special electron stained technology for studying the ultrastructure changes of basement membrane.

    To investigate the relationship between the impulse oscillometry method and plethysmogrophy used in measuring airway resistance, Using the two methods to measure the airway resistance and the ventilation data of 158 patients. The varies of resistance such as Rtot, Rocc, Z rs, R5, R20 had a high degree corelation with FEV 1, and so do the corelation among the varies of resistance. The distributions of R5 and Rtot were almost the same, but after the agreement test, there was difference between them. R5 was less than Rtot. R5 could nearly reflect the change of Rtot, but they were not, equal. The application of the impulse oscillometry method to measure the airway resistance should have different positive criteria from that of plethysmogrophy.

    To explore the clinical significance of four parameters of platelet in patients with liver cirrhosis and severe hepatitis. 521 patients with varied types of viral hepatitis were divided into 2 observing groups (liver cirrhosis and severe hepatitis) and 3 controlled groups (chronic active hepatitis, acute hepatitis and healthy adult). Four parameters of platelet (PLT, MPV, PCT and PDW) were detected (using Japan East Asia Sysmex F 800 hematocyte automatic counting machine) and compared. PLT and PCT in patients with liver cirrhosis and severe hepatitis decreased significantly (P<0.05 or P<0.01), especially in those who died or bled obviously. This suggested that the four parameters of platelet can not only reflex platelet function, but also help evaluate the severity and hemorrhage trend of viral hepatitis.

    To examine serum levels of soluble Fas(sFas) and tumor necrosis factor(TNF) in chronic heart failure and its relation to heart functional classes, 30 patients with coronary heart disease and chronic heart failure were observed. The study included 19 in NYHA heart functional Ⅲ, 11 in functional Ⅳ and 24 at the age matching normal control subjects. Serum sFas and TNF were measured by enzyme linked immunosorbent assay. Results: sFas(7.83±5.66) μg/L and TNF(5.58±1.84) μg/L were increased in CHF than that in the control group, and sFas increased with severity of heart funtional classification. The increase of sFas and TNF may play an important role in the pathogenesis of CHF.

    To investigate the effect of continuous low dosage gastrointestinal dialysis in uremic patients ,fifty-eight patients with uremia were divided into two groups at random, the treatment group in which thirty patients were given non-absorbed mannitol solution and the control group in which twenty eight patients were given Coated Aldehyde Oxystarch. After treatment, there were significant improvement of uremic symptoms, living quality and nutritional satatus,significant reductions of serum creatinine, blood urea nitrogen,serum phosphorus and uric acid, and significant increment of creatinine clearance in the treatment group. The results of the treatment group was obviously better than that of the control group (P<0.01). Either in treatment group or control group, no significant changes in blood pressure、 body weight、total plasma protein、serum albumin、proteinuria and urine amout were observed. The results showed that the effect of continuous low dosage gastrointestinal dialysis in uremic patients was obvious. It might play an important role in delaying the progression of chronic renal failure.

    To determine the role of verapamile in induction of acute heart failure, Inj. Verapamile and ten dogs were used in the experiment. The experimental procedure is as follows: ① general anesthesia and insertion of the monitoring lines, ② induction of acute heart failure with Verapamile, ③ the collection of data, ④ statisticse. The result shows that: after a satisfied induction of heart failure, there is a significant decrease in aortic systolic pressure (-29%), aortic diastolic pressure (-18.5%), +d p /d t (-47.5%), -d p /d t (57.5%), HR (-30.6%), Cardiac output (-34.2%), endocardial viability ratios (-13.8%), and coronary flow (-40.3%). However there is a significant increase in pulmonary capillary wedge pressure (70.5%). In conclusion, in the acute experiment, the administration of Verapamile is an effective mothed in animal model of heart failure.

    To explore non-operative methods and the opportunity of operative in patients with severe acute pancreatitis (SAP), 223 patients with SAP were treated in our ICU from 1990 to 1999. Non operative methods included fluid resuscitation, reduction of pancreatic secretion, elimination of inflammation mediators, supportive measures and close supervision, nutritional support, prevention and treatment of infections etc. Indication for operation in patients was pancreatic infection from 1990 to 1994.Aggressive debridement with placement of several drains into lesser sac for the purpose of continuous lavage was performed on the patient of the infectious pancreatic necrosis or fluid collection. The drainage of pancreatic abscess or localized infection with debridement of necrotic tissues was adopted from 1995 to 1999. Twenty-three patients underwent operations due to pancreatic infection during the treatment. The postoperative mortality rate of pancreatic abscess or localized infection was lower than that of infectious pancreatic necrosis or fluid collection. Active ,effective and comprehensive non operative treatment is the basic treatment of SAP. The operation of pancreatic abscess or localized infection is more effective than that of unlocalized pancreatic infection.

    To investigate the generation and possible significance of plasma trypsinogen activation peptides(TAP) in experimental rats of acute pancreatitis, 90 SD rats were randomly allocated to five groups: group EP and NP with retrograde ductal infusion of 3%and 5%sodium taurocholate ; group TP with Ulinastatin(UTI) treatment; group CP with 0.9%NS retrograde ductal infusion; group OP with sham operation. Plasma TAP was determined by EIA. Result: At times 3 h and 6 h after infusion, plasma TAP concentration of group NP (4.798±0.169、3.999±0.299 nmol/L) were significantly higher than that of group EP(2.416±0.148、3.356±0.211 nmol/L); At 6 h after infusion, plasma TAP concentration of group TP(1.611±0.113 nmol/L) was significantly lower than that of group EP. The result shows that plasma TAP concentration may be used as a marker for the early assessment of the severity of pancreatitis.

    To establish a simple,typical experimental animal model of acute necrotizing pancreatitis, rats were given 2.5 mg/g body weight of L-Arg intraperitoneal injection twice at an interval of 1h. Blood samples were collected for determination of serum amylase, and the pancreas were removed for the histological examination and the pancreatic wet/dry weight ratio delection. Result: The serum amylase of rats given L-Arg were significantly increased.The histological examination revealed that pancreas degeneration and necrosis,and the pancreatic wet/dry weight ratio were markedly increased. It shows that the administration of L-Arg overdoses seperately can induce a simple, reproductive experimental animal model of ANP.

    The aim of this study was to examine the association of cholecystectomy, gallstones and colorectal cancer. Participants of 165 had a colorectal cancer diagnosed between 1990~1999. Seven patients underwent cholecystectomy and 37 patients suffered from gallstones. Forty four colorectal cancer patients(26.7%) experienced cholocystectomy or gallstones, which are clearly more than those in the case control group(6.0%). There was significant difference between the two groups(P<0.01). Cholecystectomy and gallstones may indicate patients at high risk of colorectal cancer.

    To prove that increased bile salts cause the hepatic resticuloendothelial cell dysfunction, collagenase perfusion of the liver and isopycnic sedimentation in a two step Percoll gradient were used to isolate Kupffer cells of wistar rats. After a period of culturing, glycochenodeoxycholate(GCDC) of different terminal concentrations was added respectively. Measuring of MTT, neural red phagocytosis and concentrations of TNF in supernatants was examined to determine how GCDC affect the activity, phagocytic function and TNF release of Kupffer cells. When the concentration of GCDC was less than 25 μmol/L, GCDC had little effect on cultured Kupffer cell. But if GCDC concentration was beyond 50 μmol/L ,functions of Kupffer cells were inhibited significantly by GCDC in a concentration dependent manner. Therefore, the inhibiting function of the bile salts to Kupffer cells probably plays an important role in development of obstructive jaundice.

    To disscuss the clinical characteristics, pathogenesis and management of traumatic intraventricular hemorrhage (IVH), 50 cases of IVH were included in this study. The etiology, mechanisms, clinical and CT characteristics, management and prognosis of this injury were analyzed. Thirtytwo(64%) of the cases were caused by traffic accidents. Most patients presented meningeal irritation and fever. In 19 of the cases, the complication of primary brain stem injury was clinically diagnosed. 15 cases could be diagnosed as diffused axonal injury according to CT scan. In the group of death, Glascow Coma Scale(GCS) was significantly lower than that in the survival group, while the Graeb Scale of IVH was significantly higher. Most patients were complicated by other kinds of traumatic brain injury through CT test. In treatment, craniotomy was performed on 5 patients. V P shunts were performed on 3 patients. As the result, 24 patients died, 2 patients were in a vegtative state, 8 patients were severely disabled, 6 patients were mildly disabled and 10 patient were fully recovered. Traumatic IVH has a close relationship with diffused axonal injury. Both lesions are caused by shear stress in deep structure of the brain. The complicated injuries which need more attention in management are commonly more serious than the IVH itself. The prognosis of this injury depends mainly on the ages of patients, state of consciousness on admissison, the amount of hemorrhage, types and degrees of complicated injuries, degree of fever and vasospasm etc.
    Discussion on Clinical Results of Initial Empty-stomach Blood Sugar from Cerebral Hemorrhage within 24 Hours

    After testing the initial empty-stomach blood sugar of 216 cases of cerebral hemorrhage in their acute stages, comparison in groups was made between those of diabetic patients, those of nondiabetes with high blood suger and those of normal level of blood sugar. The results showed that the group of blood sugar disease and the group of non diabetic disease with high blood sugar are higher to the conspicuous extent than the normal group in terms of sizes of infarction. The degree of deficiency in the nervous functions and the rate of death were higher than the group of normal blood sugar(P<0.01). The results indicates that attention should be paid to testing the changes of blood sugar during the acute stages of cerebral hemorrhage and actively controlling the blood sugar.

    In order to clarify the changes in ovarian function after hysterectomy, cases having undergone hysterectomy with ovaries remaining were followed up in one to three post operative years. For the patients having undergone hysterectomy with one-side or both-side of ovaries remaining, the ovarian function is lower than normal ovarian function, and the function is declined earlier. In the third post operative year, the ovarian function decreased significantly, and was even lower than estrogen level in the early follicular phase in women of the same age. Therefore,patients with hysterectomy should be supplied with timely estrogen.
