The morphological changes of cultured bovine chromaffin cells were observed and the dopamine secreted was determined by HPLC to explore the feasibility of gene therapy with adrenal medulla cell transplantation. Results: The chromaffin cells in suspension culture, aggregated in round shape all the time and could secrete dopamine, and were still alive after resuscitation from cryopreservation; the adhesion cells showed round shape first, then stretched out short processes, turned in to shapes as triangle, fusiform, and polygon et al and restored to round shape after a week of culture. The HPLC results showed that the dopamine content arrived at the peak at about 5~6 days of culture. Adrenal chromaffin cell is a valuable source that can be used for gene therapy.

Five primate models for Parkinsonizm, which was established by 1-methyl-4-pheny-1,2,3,6-tetrahydropyridine(MPTP) injecting through unilateral internal carotid after 6 weeks, were scanned by MR imaging. -weighted images. Histological examination showed a loss of tyrosine hydroxylase immunoreactive neurons and pervasive gliosis in the ventral midbrain area on the injection side by Grino staining. The results manifested that MRI can be a modality to evaluating the primate modal of Parkinsonnism after MPTP exposure.

In order to determine the effect of the compound 962 capsule on the choline acetyltransferase and acetylcholinesterase in cerebral cortex of aged rats, and to probe the anti agedness mechanism from the view of choline energy nerve. Male and female aged rats(22-month old and 24-month old) were classified into five groups young——control group, old model group, piracetem group (0.56 g/kg), 962 middle dose group (0.9 g/kg) and 962 high dose group (1.8 g/kg). All test drug was administrated for 1~2 months by ig. The choline acetyltransferase and acetylcholinesterase activity were measured by Fonnun and hydroxylamine colorimetry method. Results: Compound 962 improved the activity of choline acetyltransferase (

< 0.05), but it had little effect on the acetylcholinesterase in cerebral cortex of aged rats. This indicated that by affecting the central nervous system in aged rats, compound 962 capsule could improve the aging process.

Twenty mice in 3 days of gestation were divided into 2 groups. Ten mice were subcutaneously injected with proteins derived from the isolated granulated metrial gland cells (as experimental group), the others with PBS (as control group). All mice were killed on the 5th day after injection. The numbers of pregnant mice and the implanted blastocysts were counted. Nine out of ten mice in experimental group were pregnant with totally 112 implanted blastocysts, whereas 7 out of ten mice with 87 implanted blastocysts in control group. The results indicate that proteins derived from the isolated GMG cells may promote the implantation of blastocyst.

The research on the effects of sigle and combined application of propranolol and nifedipine on portal hemodynamics in experimental cirrhotic rats were carried out by testing the rats′ average artery pressure, portal pressure and portal bloodstream. The results demonstrated that propranolol and nifedipine, whether used singly or in combination, could both decrease the portal pressure and rate of portal bloodstream (

<0.01) in the rats, but neicher had apparent effect on the average artery pressure, and furthermore, there were no apparent diferences on the effects between the two drugs. In conclusion, combined use of propranolol and nifedipine has a distinct improvement on portal hemodynamics in experimental cirrhotic rats, whose effect is approximately equal to either of the drugs used singly.

To report the perioperative characteristics in transmyocardial laser revascularization (TMLR) in senile patients .TMLR was performed on the beating heart with high power CO
2
laser (the Heart Laser,

PLC. Inc.)in 33 high risk cases. Transesophageal echocardiography (TEE) showed the average number of channels was 22±5. The follow up ranged from 1 to 24 months was made to assess angina class and myocardial perfusion. 2 patients died of acute myocardial infarction and one died of cardiac shock. Some people suffered related complications such as arrhythmia, pneumothorace, etal. The follow-up included 26 patients. The angina status were improved in 23(88%) patients.

Tl-SPECT showed a remarkable improvement in reversible ischemia in 16(62%) follow up patients. TMLR for patients with high risk is safe and effective. Monitor during operation is very important for prognosis.

To investigate the effects of fluvastatin on blood lipid in inhyperlipidemic model of rabbit after iliac artery endothelial denudation and the relationship between serum lipid and intimal thickening in the study, the New Zealand white rabbits were randomized into three groups: drug treatment group(

=7). The atherosclerotic lesions were developed by endothelial denudation with balloon catheter and high dosage of cholesterol diet at the same time. The experimental animals were killed and iliac arteries were removed at 1、2、4、8 weeks after endothelial injury respectively and bloodsamples were taken at the same time. By the methods of routine HE staining, blood lipid measure and image analysis system, observed the effects of fluvastatin on the level of serum TC、TG、LDL-C and the thickness of intima and media,intima/media(I/M) ratio and the relationship between serumlipid and intimal thickening. The level of TC、TG、LDL-C in drug treatment group was remarkably lower than that in non drug treatment group(

<0.05). The thickness of neointima and I/M in drug treatment group was much thinner than that in non drug treatmeant group. Coefficient of linear correlation between intimal thickness and TC、TG、LDL-C were 0.911,0.892,0.942 (

In order to assess the relationship between different HP bacteria types and their pathogenicity,investigated 1 674 patients who underwent gastroscopy and 100 blood donors. HP infection was diagnosed by at least 2 positive results from PCR, histology (Giemsa staining), or rapid urease test. Serum CagA IgG antibodies were measured by ELISA. It was found that total prevalence of CagA seropositive was 49.70% in patients and 38% in controls (

To explore the potential impact of COX 2 on cerebral ischemic injurious mechanism,the present studies examined the influence of calcium channel blocker on the expression of COX 2,which is the key enzyme in arachidonic acid metabolism. The model of middle cerebral artery occlusion (MCAO) and immunohistochemistry staining were used to compare the expression of COX 2 protein in cerebral ischemic group with that in nimotop treatment group. There were no immunochemistry positive staining cells found among either the sham operation subgroup or the subgroup after MCAO for 1 and 4 h; however, positive staining cell began to acumulate after MCAO for 12 h. The positive cells in nimotop group were significantly higher than that in group without intervention: 12 h subgroup 56±11/mm 2 vs 45±18/mm 2(

The objiective of the study is to observe the changing in risk factors and onset age of stroke in recent years. 2 154 cases of patients from 1995 to 1996 were collected, and 10 547 cases of data from review of cases in past 25 years (1972~1996) were compared. The percent of patients younger than 51 is 16.32% for 1972~1976, 22.72% for 1992~1996 respectevely. The percent age of patients younger than 41 is 1.29% 1972~1976, 7.83% for 1992~1996. Risk factors of patients are hypertension (81.1%), hyperlipoidemia(68.4%), hyperglycosemia(54.4%), and the occurrence of cerebrovascular disease tends to transit to younger group of people.

To evaluate effect of Sandostatin in prevention and treatment of stress ulceration for acute severe brain injury patients, 30 patients with acute severe brain injury (GCS 3~8) were admitted into this study. Besides neurosurgery routine treatments, they received sandostatin during the first 7 days (0.1 mg, per 8 h) and were observed in the next 3 weeks. The other 86 patients with acute severe brain injury, as the controls, just received histamine H 2 antagonist treatment. Only 4 (13.3%) of the 30 patients in sandostatin group suffered from upper gastrointestinal hemorrhage, and all happened in the first 24 h and were controlled within 1 d. In the control group, 16 patients (18.6%) suffered from upper gastrointestinal hemorrhage(

<0.05). Conclusion: sandostatin is effective for prevention and treatment of the stress ulceration in acute sever brain injury patients.

To investigate correlative factor and prevention and treatment of postpartum hemorrhage, 317 cases of postpartum hemorrhage were reviewed. The incidence of postpartum hemorrhage, during January, 1986 to June, 1999 ,were 1.53%(317/20 768). The chief reason of postpartum hemorrhage was uterine inertia, taking up 53.31%(169/317). The incidence that caesarean section occurred postpartum hemorrhage were 2.41%(151/6 278) of the cases, which was higher than vaginal delivery(

<0.01). Conclusion: Reducing the uterone inertia and caesarean seclion is the key to reduce the incidence of postpartum hemorrhage.

To study situation of growing trachea inserted with self expandable metal stents and provide the base of treating stenosis of trachea in childhood with the stents, stents of different tension being inserted into tracheas of two groups of piglets. The symptoms of the animals, the changing diameters of tracheas and stents, and the appearance of gross specimen and histopathology were observed. All, excep one, the animals had little or no symptoms. Stents inserted migrated in two animals. No trachea stenosis happened in low tension stent group. Inflammation existing in trachea of low tension stent group was milder than that of middle tension stent group. There is possibility that low tension and large diameter self expandable metal stents can be used to treat the stenosis of trachea in childhood.
