    Use of C3b-ELISA for the Detection of IK and CIC in Serum Separated by PEG Precipitation

    The purpose of this work was to detct IK and CIC in the same serum by C3b-ELISA. It was demonstrated that serum IK and CIC was separated effectively by an initial precipitation step using 3.5% PEG (M. W. 6000). IK contained in the supernatant and IC present in the solubilized precipitate were able to bind specifically to solid-phase C3b. It was worthy of note that in sera of ICD patients with either positive IK or IC. the percentage of IK and IC double positive results varied in the tests from 45% to 96%. The kinetic observation on patients with IGD suggested that there existed a parallelism between the percentage of double postive and the severity of the disease. C3b-ELISA is simple, rapid, and inexpensive to perform and it seems to provide a specific, sensitive, and versatile tool for the quantitative analysis of IK and IC.

    An assay for the specific detection of HBsAg-CIC composed of different immunoglobulins (IgG and IgM) by ELISA using anti-HBs attached on solid phase was reported in this paper. thismethod was most suitable for the detection of HBsAg-CIC with slightly exce-ssive antigen. The results of the detection in different types of patients with chronic HB showed that the incidence of HBsAg-CIC in HBAC and CAH was remarkably higher than that in CPH, in ad-dition, BHsAg/IgM was more close-ly related with the difference of disease types and with the activity of liver damage.

    Parallel detections of RF, IK and HBsAg-specific CIC were performed in different types of patients with chronic HB. Our study disclosed that the incidence of RF, IK in HBAC and CAH was remarkably higher than that in CPH, and this result was consistent with the difference of the incidence of HBsAg-CIC among these types of disease. In addition, in HBAC and CAH the incidence of RF and IK clo-sely followed by HBsAg-CIC was higher and their keeping time was also longer than those in CPH. Dynamic observations showed that there existed a consistent changes among HBsAg-CIC, RF, and the level of the comple. mentconsumed by soluble CIC in patient's sera, and a close relationship with the aggravation of hepatic func-tion impairment. The data suggested that RF and IK might be harmful pathogenetic factors to the host in chronic HBV infection.

    The present study dealt with seve-ral influencing factors in quantita-tive ELISA technique for the detec -tion of IgM-RF and IgG-RF. We selected the appropriate conditions of the experiment and standarized them. we observed and analysed the interaction between IgM-RF and IgG-RF. The result showed that IgG--RF also influenced the detec-tion of IgM-RF, i. e. IgG--RF might elevate the level of IgM-RF. It was the first time we used the internatinal standarized product of RF of WHO in our country and standarized the RF serum which would be used as a control in our laboratory with the hope that it might lay a foundation for compa-ring the results among different laboratories here after. Clinically, we detected 367 cases of patients with five diseases (RA, SLE, HB, TB and GN) with this technique. The result showed that the signi-ficance of IgG-RF was not less than that of IgM-RF in some diseases. This technique is not only simple, stable and easy to be standarized, but also sensitive and specific. It can be applied in common laboratories and is of some help to appraise the curative effect and prognosis.

    Galactose-specific Pseudomonas aeruginosa lectin was successfully purified by affinity chromatography on D-galactose-starch gel, which was prepared by using cross-linking reagent epichlorohydrim, and the results involving its yield, molecular weight and amino acid composition were compared with those obtained by affinity chromatography on Sepharose 4B. The optimal conditions for purification with D-galactose-starch gel method were also observed.

    Galactose-sepecific lectin from pseudomonas aeruginosa could agglutinate the erythrocytes of various animals (including rat, rabbit, mouse, sheep and pig) and the ability to agglutinate the erythrocytes treated with papain was stronger than those untreated except erythrocyte of chicken. The hemagglutination activity of lectin was inhibited by galactose, N-acetyl-galactosamine, lactose, raffinose, melibiose, xylosa and Na-EDTA, but not by glucose and N-acetylglu-cosamine. Binding of galactose to the lectin was assayed spectro-photometrically and the association constant for galactose was 6×103L/ Mol. In addition, gel-filtration analysis revealed that the monomer of the lectin exhibited a tendency of aggregation.

    A Preliminory observation and mechanism survey about the effect of immune regulator-BCG on the capacity of resistance against pseudomonas aeruginosa (PA) infection in mice were reported in the present paper. The mice preinfected with BCG (1mg/per mouse iv) were shown to be highly resistant to subsequent challenge with PA (3LD50), and susceptible to lethal effect of rough PA endotoxin. The results of our experiment showed that BCG-induced resistance to PA was related to an enhanced eliminating ability produced by fixed phagocytes of mice and BCG-induced susceptibility to PA endotoxin might be attributed to blockage of reticulo-endothelial system of mice by BCG. The relationship of experimental results was discussed.

    The neutralization of F (ab')2 fragment of anti-HCG-IgG with HCG was investigated using mouse uterine weight assay and uterine histological examination. No significant increase in uterine weight and endometrial hyperplasia were observed in low dilution of F (ab')2 fragment of anti-HCG-IgG. The endometrial hyperplsia was not found on histological section, but in relatively high dilution, a serious uterine edema and hyperemia with infiltration of phagocytes occurred. These investigations show that the Fc portion of anti-HCG is not roquired in the neutralization with HCG.

    A new solid phase-dish coated with polystyren using sandwich ELISA to determine HBsAg was described in this paper. The results of dish-ELIS A and its sensitivity were compared with conventional polystyrene microtitration plate ELISA on 170 patients serum specimens. No significant difference was observed. This method was able to detect as little as 0.005 ug/ml of HBsAg. The advantages of dish-ELISA including sensitivity, simplicity, speediness and low cost render this solid phase suitable for ELISA.

    Myoglobin is found exclusively in striated muscle. It can be considered a fetal antigen. Myoglobin was prepared from normal human thoracic muscle and was used to immunize the rabbits to obtain antimyoglobin antisera. The indirecf immunoperoxidase reaction was performed on numerous normal tissues and a variety of tumors. All the tissues were paraffin-embedded and formalin-fixed. The resuits from skeletal and cardiac muscle and rhabdomyosa-rcoma were positive. Of six cases of rhabdomyosarcoma, five were positive; one was undefined. Therefore, myoglobin should be considered as a specific marker for striated muscle and rhabdomyosarcoma, and the immunoperoxidase technique may be a useful method for the diagnosis of rhabdomyosarcom and other neoplasamas with rhabdomyoblastic differentiation.
