Measurement of Sodium Pump Activity in Sheep Cardiac Purkinje Fibres

Raising the internal sodium ion activity (a_Naⁱ) by way of intermittent fast beating the sheep cardiac Purkinje fibers in different frequencies resulted in declining of a_Naⁱ at the very end of the train of beats.Both of a_Naⁱ increments and declining rates (d(a_Naⁱ/dt) paralleled with the stimulation frequencies.The values of d(a_Naⁱ)/dt were plotted against the corre-sponding a_Naⁱ to obtain a correlation curve (sodium pump-internal sodium relationship, SP-IS).Sodium pump activity can be evaluated by comparing the slopes of correlation curves.The intersects of the correlation curves and the ordinate (Vmax) was got with extrapola-tion.The sum of values of Vmax and the maximum d(a_Naⁱ)/dt represented the maximum ac-tivity of sodium pump.The pump activity can be calculated simply and repeatedly with this method.The effects of high/low pH, K⁺, Ca²⁺, and 4 mmol/L Mn²⁺, 5×10^-7mol/L iso-prenaline on pump activity were observed simultaneously.

The effect of MPS on anti-SRbc antibody forming cells in mice was tested by haemoly-tic plaque method.The result showed that MPS may enhance the antibody production in mice spleen cells as compared with the controls(P<0.001).It indicated that MPS might enhance the resistant ability of patients with infective desease and increase the antibody response after immunization.

For enhancing the purification recovery of carboxypeptidase B, the Felk’ s method was modified as following: (1)Both of Tris-HCl buffers with contained 0.1 mol/L and 0.15 mol/L NaCl were ap-plied respectively in the batch adsorption and elution from DEAE-cellulose.(2)In the second DEAE-cellulose chromatography, the DEAE-cellulose column was e-luted with linear gradient of NaCl from 0.00 to 0.09 mol/L in Tris-HAc buffer.(3)Carboxypeptidase B was eluted completely with 0.09 mol/L NaCl in Tris-HAc buffer.The purity and the molecular weight were identified by TLC scanning method after gel electrophoresis.The results showed that recovery of enzyme activity was in creased from 8% to 15%, specific activity was increased from 175 U/mg to 192 U/mg and purity of en-zyme was reached 91%.

An animal model of the femoral artery thrombosis was produced in rabbits.Preliminary investigation was made in 60 models rabbits to observe the characteristics of the morphotogic change during the thrombosis process from 1 to 6 days as a contral.Mai Tong Ling No.2 was administered through gastric tube in 18 animal models (thrombus of 3rd day) which was divided into three groups (with different time period of medication).The morphologic change of the thrombus was observed after medication to evalute the thrombolytic effect of Mai Tong Ling No.2 on the process of thrombosis.

The determination of rutin content was studied with double-wavelength technique to e-liminate the disturbance of quercetin.When the concentration of quercetin attain to 20 mg/L, the error rate was not beyond 0.4%.The recovery rate was 94.2% to 100.2%.

Angioarchiteture of the CNS were visualized with peroxidase cytochemistry in rat.This technique can successfully demonstrat that the endogenous peroxidase activity of red cells and granular leukocytes(especilly polymorphonuclear and eosinophilic)were excellent mar-kers for revealing CNS blood vessels at the light microscopic level when 3,3′-diaminobenzi-dine (DAB), benzidine dihydrochloride (BDHC), O-dianisidine (O-D) and tetramethyl-benidine(TMB) were used as chromogen.The four peroxidase cytochemical approches are useful for visualizing CNS microvasculature in nomal laboratory animals; the four approches have potential application to cerebrovascular studies of stroke, trauma, neoplasia and intra-cerebral grafts.

The serum antibodies to HLA-A,B,C and DR which are on T and B lymphatic cells were examined by complement-depending standard method of NIH, which serum were tak-en from 7 patients who had received intracephalic transplantation of fetal adrenal medullary tissue.Every serum sample was mixed with T and B lymphocytes of 16 normal persons to perform micro-cytoxin reaction.The result presented postitive HLA-DR reaction in the serum from a patient.It showed that the rejection should be on regard in engaging intra-cephalic transplantation of fetal tissue clinically and should give more attention to immunore-jection.

It is sure that BNP plays a important role in regulation of aldosterone (Ald) secretion, but no one has reported that cerebral BNP (cBNP) and peripheral BNP (pBNP) have any difference in its regulation action.In order to solve this question, we have compared effects of cBNP administrated by icv and pBNP given by iv alone or associated with angiotensin II (AT-II), Adrenocorticotro-pin (ACTH) and Arginine Vasopressin (AVP) on regulation of Ald secretion.The results show that 1) AT-II, ACTHand AVP given by iv (5 μg/3ml· h^-1) could increase the concentration of plasma Aid.BNP (5μg/3ml· h^-1) inhibited the stimulation of ACTH and AVP, although BNP had not been discoved to have effect for AT-II; 2) BNP (2/μg in 10/μl NS)administrated by icy decreased the concentration of plas-ma Aid.icy injection of AVP (2 μg in 10 μl NS)could increase plasma Aid, but ACTH (2 μg in 10 μl NS) and AT-II (2 μg in 10 μl NS)did not have this effect.However, giving ACTH (2 μg in 10 μlNS) or AT-II(2 μg in 10 μl NS) individually with BNP (2 μg in 10 μl NS)could induce paradoxical stimulation on plasma Aid concentration, and 3) there were no effect on stimulation of Aid secretion induced by iv perfusion of AT-II, ACTH or AVP (5 μg/3ml· h^-1) when BNP (2 μg in 10 μl NS) was injected by icy.From above results, we found that there are certain differences between cBNP and pBNP on regulation of Aid secre-tion.Cerebral ACTH and AT-II can induce the increase of plasma Aid only in the condition of coo-peration with cBNP.

Laparoscopic cholecystectomies are successfully performed by us for 24 cases.It is a new, safe and effective operation.The benefits of laparoscopic cholecystectomy are minimal trauma; minimal pain; and quicker recovery.To avoid the complications; such as bile duct injury, this report describes someimportant things in the patient selection; anesthesia moni-tor; patient’s position; the trocar site; the exposure of the operation field; the anatomic dis-section; the removal of gallbladder and the training of surgeons, our new technic is a deve-loped surgical procedure.
