Preliminary Analyses on the Soluble Proteins and Antigens of Alveolar Hydatid Tissues and Cyst Fluid

The soluble proteins of cyst fluid,hydatid cysts,protoscoleces and cyst walls of alveolar hydatid obtained from experimentally infected gerbils(Meriones unguiculatus)were studied with SDS-PAGE.Four specimens showed respectively 23,26,26,and 25 protein bands with their major bands being respectively 4,7,5,and 7.Four kinds of soluble proteins mentioned above were tested as antigens with Dot-ELISA.Asensitivity of 67%,67%,100%, and 67% were found while the specificity showed 90%,89%,83.5%,and 91% respectively.

The dot enzyme-linked immunosorbent assay(Dot-ELISA)and standard enzyme-linked immunosorbent assay(ELISA)were used to detect Cysticercus cellulosae antibodies in the sera of the patients with cerebroeysticercosis.By using Dot-ELISA,47 out of 52 pa-tients(92.3%)showed a positive reaction.In the standard ELISA,47 out of 52 patients (90.4%)showed a positive reaction.None of the 40 serum samples from normal controls was found positive by both assays.Although there were similar higher sensitivity and speci-ficity in the diagnosis of cerebrocysticereosis by both Dot-ELISA and standard ELISA,the Dot-ELISA is more economical,simple and do not need any specific equipment for the diagnosis of cerebrocysticercosis.Therefore,Dot-ELISA is suitable for clinic application and epidemic survey.

40 adult male rats were used and divided into 4 group,i.e.normal control group, “spleen deficiency gruop”caused by rhubarb,spontaneons recovery group and therapeutic group with Chinese herbs(Si Jun Zi decoction).The stomach of the four groups were removed and processed for demonstration of the reactivities of succinate dehydrogenase (SDH),Lactic dehydrogenase(LDH),magnesiu m-activa ted adenosine triphoaphatase(Mg²⁺-ATPase),alkaline phosphatase(ALP),non-specific esterases(NSE)and carbonic anhydrase (CA).The reactivities of above mentioned enzymes in gastric mucosa were observed with light microscopy and quantitative analyses were performed.The present study revealed that the reactivities of SDH,CA and NSE in parietal cells of fundic glands in“experimental spleen deficiency group”were less than those of the normal control group,and LDH reactivity was elevated as compared with the normal control group.These enzyme activitied of spontaneous recovery group appeared similar to them of the“experimental spleen deficiency”group.The enzyme activities of the therapeutic group seemed to be similar to the normal control group.The activities of Mg²⁺-ATPase and ALP in all the four group did not show any difference.Our study suggested that these changes of enzyme activities may be involved in pathogeneses of digestive disorders presented in spleen deficiency syndrome and the Chinese herbs(Si Jun Zi decoction)possessed significant therapeutic effect.

Rheumatoid factor(RF)was detected on synovialis of 20 patients with rheumatoid arthritis by immunohistochemical method.IgM RF and IgG RF positive rate was 60% and 45% respectively.According to the clinical laboratory tests and pathological examination,the detection of RF on synovialis is considered as an useful diagnosis for rheumatoid arthritis in same specimen,especially as a crucial test to except the other inflammative arthritis in the application of arthcope.

A new kind of iron-supplementary drug——iron(Ⅲ)-tremella fuciformis polysaccha-ride complex(ITC)was synthesized with FeCl₃ and tremella fuciformis.Because it was not free Fe³⁺in the ITCsolution,therefore this complex was very stabe.The investigation by infrared spectra,ultracentrifugation and electron microscopy showed that structure of ITCwas an interracial compex with β-(FeOOH)_n.core and a high polymer.The diameters of complex particles ranged from 30～80 nm.Because it was not precipitated and dissociated at the physiological pH it higher bioavailability would be expected.

Biotin-labeled DHA probes of human papillomavirus 11,16,18,51 types are produced by the following procedures.Fitst,culture of the strains of E.Coli DH-52 which contain cloned puc-18 plasmid and plasmid amplification.Second,extraction and purification of the plasmids after alkaline denaturation.Third,enzyme digestion of the plasmids.Fourth,biotin-labeling of HPV 11,16,18,51 DNA probes.The sensitivity of HPV 11,16,18,51 DNA probes.is detected by using dot hybridization.The results is that it is 0.05 pg for HPV 11,16,0.25 pg for HPV 18,0.1 pg for HPV 51.

We use TCM Knowledge Representation Language——TCMKRL and corresponding Expert System Generating Environment——TCM-ES-GE to develop famous veteran TCM doctor Liang Zonghan's expert system of pediatrics disharmony between spleen and stomach.The prescriptions given by the system agree with Dr.Liang's thinking rules.The system has friendly user interface.It is convenient to maintain and extend the system.The practice indicates that it marks a new advance that TCM doctors can make up expert systems using tools independently.

It will be benefited with solving knowledge acquisition bottle problem in developing expert systems.It can be mastered and used by TCM doctors.Expert system generator developed is a powerful software tool designed according to computer-aided software engineering (CASE)principles.It is possible to use it to develop TCM expert systems in batches.

Twenty-seven specimens of adaut salivary glands were studied with five histochemical methods.In these cases,26 were normal and the other one was chronic inflammation.The result shows that the secreting substances of the similar mucous cells are quite different in histochemistry,even in the same specimen,the mucous substances were various. Therefore,it is thought that there are some limits in using the morphologic classification in salivary glands.It is not exactly correct to think the parotid as a pure serous gland because there are a few mucus substances in the parotid secreting cells.On the other hand,when there is a chronic inflammation of salivary gland,the ductal mucous cells were hyperplasia, and the mucus from the duct mucous cells was not only increased in quantity but also changed in its quality.

The components of soluble antigen of Paragonimus vestermani(PwA)were analysed with immuno-electrophoresis(IE),Sodium dodecyl sulfate-polyacrylamide gel eleetrophoresis (SDS-PAGE),and western blot.Two broad and distinct bands only were detected in IE. 29 protein bands were found to be present when adult worm homogenates were subjected to SDS-PAGE analysis,and their molecular weights ranged from 90.0～6.0 kD.The molecular weight of the 10 most prominent protein bands was corresponeded to 57,53,49.5,46, 42,30,27,25,14 and 13 kD respectively.The western blot examination of PwA with sera of proven cases of paragonimiasis revealed 3 bands,whose protein molecular weights were 25, 13,and 8 kD respectively.

Blymphocytoid cell lines of 8 patients with diabetes and 2 Control have been studied.It was found that the ultrastructure of Blymphocytoid cell line from patients with diabetes, was different from PHA-stimulated lymphocytoid cell and Burkitt's lymphomd cells,but it was similar to EBV-carring diploid lymphoblastoid cells in literature published.Significant pleomorphism,more active cell division,proliferation,motility and phagocytosis have been observed in these cell lines.
