HPLC Analysis of TXB₂ Content in Platelet and the Anti-platelet Actions of Acathopanax Senticosus

TXB₂ content in platelet were analysed by high-performance liquid chromatography (HPLC) using a pre-column derivatization method,in which a fluorescentreagent,4-bro-momethyl-7-methoxycoumarin (BrMmc) was used.The mothod was applied to studing the effects of Acanthopanax Senticosus (A.S.) extract,a Chinese medicine,on platelet func-tion.The platelet TXB₂ were extracted by a Sep-Pak C₁₈ (waters).In the presence of 18-crown-6 and K₂CO₃,BrMmc reacted with carboxylic group(—COOH) of TXB₂ to form a fluorescent ester (BrMmc-TXB₂),which was seperated on a column (250mm×4.6mm I.D.)packed with Zorbax-ODS (6 μm,Dupont Instruments)and monitored by a fluorescent detector in Ex 345 nm and Em 405 nm.The mobile phase (acetonitrile:water:phosphoric acid;60:40:0.1 V/V),flowed at 1 ml/min at 30℃.The seperation of BrMmc-TXB₂ was completed in 20min.The detection limit was 15 ng using the external standard method.The anti-platelet actions of A.S.in rabbit were studied.The resultes (in vitro) showed that the platelet aggregations induced by AA and ADP were potently inhibited by A.S.in a dose-de-pendent manner and the AA-induced TXB₂ formation in platelet was also reduced.After in-travenous injection of A.S.(120mg/kg),the platelet aggregations induced by AA and ADP were inhibited and the effects lasted 30 and 45 min,respectively.

The primordial germ cells (PGCs) of mouse embryo could be divided into three kinds by their size.It's defined that middle and small PGCs start increasing in two or three days. In vitro culture it looks like big PGCs.PGCs grow suspended in the 6th or 7th days.It's unknown that PGCs in vitro can propagate (reproduce).Somatic cells can grow luxumantly on the wall.Generally it dies at the same time as PGCs.

12 intraeranial Primary Germinomas were studied by immunohistochemical Avidin-Biotin-Complex method.6 of 12 cases showed positive cells of pregnancy-specific betal-glyco-protein (SP₁);3 of 12 cases were postive for human ehorionic gonadotropin (hCG) and 3 of 12 cases were postive of human placental lactogen (hPL).These cells resembled the extrav-ilous trophoblasts.According to the findings we consider the origins of intraeranial Primary Germinoma are primordial germ cells.They are able to secrete hCG,hPL and SP₁.

Mai Tong Ling Ⅰ、Ⅱ are prescriptions of Chinese herbal medicine which is effective to vasculitis,made by Beijing Xuanwu Hospital of Chinese Traditional medicine.We carried out the acutetoxicity,accumulative toxicity test and mutagenic assays of Mai Tong Ling Ⅰ、Ⅱ. The results showed that the LD₅₀ by the oral route in mice and in rate were both more than 100 ml/kg.The accumulative effect was vory low.Micronucleus test of mice bonemarrow, Ames test and analysis of spermary chromosome aberration in mice testes were all negative, mutagemicity wasn't discovered.Thus,we think that Mai Tong Ling Ⅰ、Ⅱ are relatively safe,it may be used to treat vasculitis

Chitosan has the property of binding heavy metal ions.Its maximum amount of adsorption and equilibrium time have been studied.There is no significant difference in the adsorp-tion ability between chitosan treated with nitrogen and those without nitrogen treatment. Chitosan shows nonspecific adsorption in the binding heavy metal ions such as plumbum, chromium,Copper,Cadminm.

Polyxenically cultivated Entamoeba histolytica trophozoites(HB₁ and HT₁ strains)were inoculated into the cecum of the clawed jirds (Meriones unguiculatus) after laparotomy.The histopathological changes occurred in the colon,liver,spleen and mesenteric lymph nodes of the jirds were studied from 7 to 20 days postinoculation.Of 16 inoculated jirds,12 were infected.Seven of the infected jirds showed early invasive lesions with superficial ulcerations and lymphocytic and giant cell infiltrations in every layer of the intestinal wall.Small amebic liver abscess and small focal necrosis in the mesenteric lymph nodes were found in one jird.

The livers of 30 autopsies of cor pulmonale were observed morphorlogically.Hepatic congestion,fatty degeneration,atrophy of hepatic cells,detachment of hepatic plate and the focal necrosis of hepatic cells were the main features among the findings.An acute necrosis of the lobular central hepatic cells resulting from hypoxia appeared in four patients with serious hypoxemia lasting a week.The mechanism of this hepatic cell necrosis was discussed.The author conclude that the patient with cor pulmonale may suffer from cirrhosis,if the heart function is insufficient for a long time.This kind of cirrhosis has the following three characteristics:1.With a long history of heart failure,usually more than 10 years.2.Cirrhosis could follow cot pulmonale,but much less often than other kind of heart diseases. 3.And it is characterized by abnormal lobulation.

In the present study,we report the results of endoscopic microwave papillotomy (EMPT) using Chinamade Microwave Therapewtic Instrument.EMPT was performed in 50 cases:including choledocholithiasis papillary stenosis etc.The technique was successful in all cases.The success rate of removing stones was 96%.Arelief rate of the stenosis state was 90%.Tere were no serious complications such as hemorrhage and perforation.There was no death case in our group during treatment.Also,EMPT had several advantages compared with standard EPT in terms of its cost and simplicity of the technique.

Atota of 10401 newborns were delivered from January 1984 to December 1988.Among them 2784 cases(26.77%) had cord abnormalities,which caused 31 perinatal deaths and were the leading cause of perinatal mortality.True knot and torsion of cord are cause by occasional factors and difficult to prevent.Short cord might be the result of oligohydramnios and decreased fetal movement,and it markedly increases the occurrence of fetal distress and neonatal asphyxia.Fetal death could be caused by cord entanglement with tension especially in relative short cord.As soon as prolapse of cord is diagnosed,delivery should be completed immediately.Intensive monitoring the volume of amniotic fluid and fetal movement,ultrasound scanning fetal neck to look for entangled cord in antenatal care and continuous monitoring of fetal heart rate during labor are helpful for early diagnosis.

10 patients with hypertension without complication were treated with 10 mg of nifedipine (Chinese product) three times a day.It was found that on 3rd and 7th days the blood pressure decreased on the average from 161.4/92.8 to 139.8/84.6 and 134.2/84.6 mmHg respectively;mean arterial blood pressure decreased on the average from 114.2 to 104.2 and 102.0 mmHg;total peripheral resistance decreased on the average from 1741 to 1514 and 1263 dyn·s/cm⁵ respectively.With echocardiography we found that early diastolic time-velocity integral increased on the average from 7.2 to 8.4 and 8.7 at third and seventh day.Early diastolic and atrial contraction peak flow velocity and cardiac index were also improved.From above-mentioned findings we are of the opinion that Nifedipine (produced in China) is effective in reducing blood pressure and improving left ventricular performance.

The Se,AI,Zn,Cu,Fe,Ca,Mg,P and As contents in the semum of healthy individuals and patient troups of Gastric Cancer,Atrophic Gastritis,Gastric Ulcer and Duodenal Bulbar Ulcer were determined by the Inductively Coupled Plasma Spectrometry.The results showed that the serum Se and AIlevels in all patients were lower than in normal subjects,the diferences being very significant.The Zn content in the serum of the Gastric Cancer group was lower than that of normal subjects,while the Cu content and Cu/Zn ratio were higher.The serum Fe level in Gastric Cancer was significantly lower than in the healthy controls,the Duodenal Bulbar Ulcer group and the Gastric Ulcer group.
