    Study of the Relation between Human Papillomavirus and Condyloma Acuminata with Electron Microscopy and Immuno-electron Microscopy

    30 cases of condyloma acuminata were studied with electron microscopy and immuno-electronmicroscopy. Ultrastructral changes of HPV infection(koilocytosis, binucleation, giant irregular nuclei, electron dense matrix and inoreased number of swollen mitochondria) were present in high prevalence in condyloma acuminata. But no mature HPV particles were found. The koilocytosis may result from rupturing of swollen mitochondria and worn-out cytoplasmic dense matrix. Expression of HPV antigen was shown in 60% cases with gold-staining immuno-etectronmicroscopy, and in 47% cases with enzyme-staining immuno-electronmicroscopy. Therefore, the former method proved to be more sensitive than the latter one. The majority of HPV antigen possitive cells were located in the stratum granulosum and the stratum corneum.

    In situ hybridization is a valuable method for the demonstration of the presence, localization and distribution of viral DNAS within frozen histological sections or paraffin embedded sections. Using biotin-lablled HPV6/11 DNA and HPV16/18 DNA as prodes, we studied specimens from condyloma acuminata and cervical carcinoma with in situ hybridization method. It has been shown that most of the condyloma acuminata specimens contained HPV6/11 DNA So the etiologic factor of condyloma acuminata may be HPV6/11. The HPV 6/11 DNA positive nuclei mainly located in the upper part of the epithelium, with some scattering in the deeper part. This suggests that HPV6/11 may first infect deeper part of the epithelium and the replication of viral DNA is completed in the upper part. A few condyloma acuminata patients appeared to have mixed infection, containing both HPV6/11 DNA and HPV16/18 DNA. These patients were probably at the risk of progression to carcinoma. With regard to cervical carcinoma, a high proportion of these patients contained HPV16/18 DNA. This suggests that the major etiologic factor of cervical carcinoma may be HPV 16/18. Some patients infected with both HPV 16/18 and HPV6/11. In a few cases, HPV 16/18 DNA were not found, there should be other factors responsible for the malignancy of this kind of patients.

    Structural polypeptides of human papillomavirus (HPV) isolated from common and plantar warts were analyzed by using gel electrophoresis and their molecular weights were measured. The electrophoretic analysis results of HPV indicate that there are different polypeptides patterns and molecular weights between verrucae vulgaris and plantaris.

    Successful transformation have been mede by using the recombinant plasmid of human papillomavirus type11 DNA(HPV11DNA) and pBR322 plasmid vector with E coli C600 as the receptor bacteria. Sixty-one strains were obtained and fifteen strains were selected randomly for further identification. The pBR322-HPV11DNA plasmid were isolated by both the alkaline denature method and the clear cleavage method and cut by BamHI. Then the agarose gel electrophoresis has been done. The molecular weight of HPV11DNA was assayed by compairing with the reference DNA of standard bacteriophage which was cleaved with HindⅢ. The results showed there was the DNA band of HPV11 DNA with the MW of approximately 5.5×106dts and 8.1kb. So it is convenient for the further amplification of the pure HPV11DNA and production of the genetic probe for exploring the association of the HPV11 infection with the development of sequamous cancers.

    Southern blot analysis of human papillomavirus genome was carried out on 33 cases of human cervical specimens histologically diagnosed as cervical cacinoma (26 cases), condyloma acuminatum (1 case), vulvar carcinoma (1 case) and chronic inflammation (4 cases). 69% of cervical carcinoma were found to be positive for HPV DNA. Among them 23% (6/26) were positive for HPV 16,7.6% (2/26) were positive for HPV 18 and 11.5% (3/26) were positive for HPV X1.

    Ultrastructure of eleven common warts and two plantar warts were studied by transmitional electron microscopy. HPV antigens were localizied by immunohistochemical staining. Results showed that there was no viral particals or antigen in the stratum basale and lower or middle stratum spinosum. The viruses and antigens were mainly found in stratum granulosum and corneum, sometimes in upper stratum spinusum. "Pre-virus particlesn" were seen in upper stratum spinusum. A morphogenetic course of HPV in squamous epithelial cells was discovered.

    Four specimens of colon-rectal carcinomas were stained histochemically and immunochemically using horseradish peroxidase labeled anti-CEA Fab antibody for the detection of CEA tumor marker, The results proved that Fab antibody fragment labeled with horseradish peroxidase better penetrated into the tissue owing to its smaller molecular weight, In addition, this method is simple, reliable and may be used for the detection of gastrointestinal adenocarcinomas as auxiliary procedure for pathological diagnosis.

    In the investigation of the effects of the seereta of human adenocarcinoma HT-29 cell line on mouse kidney fibroblasts, we identified that the extract of the cultured, HT-29 cell line in serum-free medium induced not only morpho-logical changes of fibroblasts in vitro, but also caused very longer survival time of fibroblasts than control group. Analysis of the extract with polyacry-lamide gel electrophoresis and chromatography revealed that it contained 3 peptides. The moleoular weight of each paptide is approximately 38904、25704 and 15311 respectively.

    In the experiment of induction of transformation of host stroma cells in nude mice by transplanted human colonic adenocarcinoma HT-29 cell liner, we discovered accidentally that the serial passage of the HT-29 cell line in nude mice caused the differentiation of human tumor cells. There appeared a lot of granular cavities, mucus-likem material secreted by differentiated adenocarcinoma cells and rich connective tissue in the sections of 3rd and 4th passage. Moreover, colonic villi-like structures can be seen in sections of 5th passage. We also found that the amount of carcinoembryonic antigen(CEA) per unit volume of tumor in serum of tumor bearing animals decreased as the number of passage was incerased.

    There were the inert IUD on the right uteru of the rats and without IUD on the left. Histochemical changes of the endometrium of the rats with IUD as follows: the activity of Gly and AKP increased, but G6Pase, NSE and SDH decreased in the endometrium of the rats with IUD during the estrus and the 6th day of pregnancy. 3-β-HSDH, ACP, and Lipid increased during the estrus. 3-β-HSDH, ACP, Lipid decreased during the 6th day of pregnancy. These results suggested that IUD had influenced normal metabolism of endometrium of the rats with IUD. It may be the mechanism for IUD to impede the implantation

    In an effort to ascertain the prevalence of obesity and thiness in young children, 3070 healthy pupils, age 6~12 years, were surveyed. The family income, the extent of parenl educated and the childrens'eredit in the school showed no siganificant difference among obese and thin pupils. These data indicate that the prevalance rate of obesity and thinness were 5.8 percent and 4.2 percent separately.

    The establishment and growth of a cell line(CT-188) from fresh incubated larvae of Culex tritaeniorhynchus in a modified 199-medium is described. Only ethanol (70%)were used for sterilizing rather than the complex of mercuric chloride(HgCl2) and ethanol. The majority of CT-188 cells were a fibroblast-type (approximately 95%); however, a small number of epithelial-like cells and other cells were also present in the culture. 10~11 days after the primary cells had covered the bottom of the container and could be subcultured. After seeding cells began to attach to the glass surface of the container within 30 minutes and reached the division peak in 48 hours. Most of the cells had the normal 2n chromosome complement of 6, although on occasion cells with more this number were seen. The cells are stored in liquid nitrogen(-196℃)at various passage levels and have been successfully resuscitated.
