In the present experiment, microelectrode was used to explore the physiological properties of the Mauthner cell with the stimuli of the skin.Direct stimulation of the skin evoked postsynaptic potentials (PSPs), which were stimulus strength-and frequency-dependent within some limits. The intracellular skin responses could be increased by passing hyperpolarizing currents to the M-cell through the Cl- filled electrode. The results suggest that the afferent pathways from skin to M-cells contain both excited and inhibitory compotents, which interact each other and modulate the excitability of the M-cells together.

The aim was to test the hypothesis that endothelial dysfunction is already evident in normoalbuminuric with type 2 diabetes.Previous studies have shown that the development of microalbuminuria is associated with the onset of endothelial dysfunction in individuals with type 2 diabetes.Plasma soluble intercellular adhesion molecule(sICAM-1), soluble vascular cell adhesion molecule(sVCAM-1), vWF were measured by ELISA in 28 healthy control subjects(groupⅠ), 57 normoalbuminuria individuals with type 2 diabetes(groupⅡ), and 25 microalbuminuria individuals with type 2 diabetes(groupⅢ).Plasma sICAM-1 levels were significantly higher in the groupⅡ and groupⅢ than that in the groupⅠ(P<0.05).In contrast plasma sVCAM-1 and vWF levels were elevated only in the groupⅢ compared with groupⅡ and groupⅠ(P<0.05).Correlated analysis showed a significant positive correlation between sICAM-1 and blood pressure, triglyceride, vWF, sVCAM-1.The results suggest that plasma sICAM-1 levels might be implicated in the development of diabetic vasculopathy, and could predict the onset of endothelial dysfunction instead of plasma vWF levels. The measurement of plasma sICAM-1 levels in type 2 diabetic patients may be clinically useful for early discovering of endothelial dysfunction.

The aim was to assess complement dependent cytotoxicity and change of IL-1 in hyperacute rejection of pig-to-human xenotransplantation.Cultural porcine vascular endothelial cells (PVECs) and human sera were used to set up an vitro model of hyperacute rejection of pig-to-human xenotransplantation.Various serums were divided into four groups: HS, HI HS, C1q-D, PS.Every serum was incubated with cultural primary porcine endothelial cells from 0.5 to 4 h, then supernatant fraction was removed to test LDH content, finally percent cytotoxicity was calculated.Morever, IL-1β in supernatant fraction was determined by ELISA.Result: 1) In the same time point, percent cytolysis of HS was higher significantly than that of control (P<0.01) and HI HS, C1q-D's percent cytolysis was similar to control's (P>0.05).As time prolonged, percent cytolysis of HS continued to increase ( P< 0.01).However, percent cytolysis of every group didn't increase after 3~4 h.2) In the group of HS, the contents of IL-1β were higher than those of control ( P< 0.01).Other groups' contents were similar to those of control ( P> 0.05).When reaction time prolonged, content in group HS went up and remained stable when reaction time arrived at 3~4 h.The results suggest that endothelial cells have an important role in the thrombosis in hyperacute rejection of pig-to-human xenotransplantation by influencing IL-1.
